Integrin ITGAM/ITGB2 is implicated in various adhesive interactions of monocytes, macrophages and granulocytes as well as in mediating the uptake of complement-coated particles and pathogens (PubMed:20008295, PubMed:9558116). It is identical with CR-3, the receptor for the iC3b fragment of the third complement component. It probably recognizes the R-G-D peptide in C3b. Integrin ITGAM/ITGB2 is also a receptor for fibrinogen, factor X and ICAM1. It recognizes P1 and P2 peptides of fibrinogen gamma chain. Regulates neutrophil migration (PubMed:28807980). In association with beta subunit ITGB2/CD18, required for CD177-PRTN3-mediated activation of TNF primed neutrophils (PubMed:21193407). May regulate phagocytosis-induced apoptosis in extravasated neutrophils (By similarity). May play a role in mast cell development (By similarity). Required with TYROBP/DAP12 in microglia to control production of microglial superoxide ions which promote the neuronal apoptosis that occurs during brain development (By similarity).

Macrophage Marker (CD11b, CD68, CD163, CD14, CD16) Antibody Panel - Human (ab254013) is a panel of 6 rabbit recombinant monoclonal antibodies to macrophage markers. Macrophage markers are proteins used to identify and study macrophages, immune cells involved in phagocytosis, inflammation and tissue repair.
This panel contains antibodies to CD11b (UniProt: P11215), CD68 (UniProt: P34810), CD163 (UniProt: Q2VLH6), CD14 (UniProt: P08571) and CD16 (UniProt: P08637) in convenient trial sizes (20 µL) to allow for efficient and cost-effect testing of the antibodies simultaneously.

Activity summary

CD11b CD14 CD16 CD68 and CD163 are important macrophage markers. CD11b also known as integrin alpha M has a mass of about 170 kDa is found on the surface of macrophages neutrophils and some T cells. It plays a mechanical role in cell adhesion migration and phagocytosis. CD14 acts as a co-receptor for the detection of bacterial lipopolysaccharides and is expressed on monocytes and macrophages. CD16 or FcγRIII is involved in antibody-dependent cellular cytotoxicity (ADCC) found on natural killer cells macrophages and some granulocytes. CD68 a marker of macrophage lineage is important in phagocytosis and is present in tissue macrophages. CD163 known for scavenging hemoglobin-haptoglobin complexes is expressed on monocytes and macrophages aiding in anti-inflammatory responses.

Biological function summary

These macrophage markers participate in essential immune system functions. CD11b binds to iC3b opsonized particles facilitating phagocytosis and inflammatory signaling. CD14 acting with TLR4 triggers innate immune responses against pathogens. CD16 engages with the Fc region of IgG antibodies mediating ADCC. CD68 is part of the lysosomal/endosomal membrane associating with scavenger receptors in phagocytosis and tissue remodeling. CD163 modulates anti-inflammatory processes by binding to hemoglobin-haptoglobin complexes leading to the release of anti-inflammatory cytokines. These markers are vital to immune surveillance and the clearance of pathogens and debris.

Pathways

CD11b and CD14 play roles in the Toll-like receptor (TLR) signaling pathway which is critical for initiating immune responses. CD11b interacts with complement component iC3b aiding in immune complex clearance. CD16 is involved in the FcγRIIB-mediated inhibitory pathway balancing immune activation. CD68 contributes to pathways involved in lysosomal degradation processes. CD163 is linked to the IL-10-mediated anti-inflammatory signaling pathway. These macrophage markers contribute to the coordination of host defense and inflammatory resolution.

Associated diseases and disorders

CD11b is associated with chronic inflammatory diseases such as rheumatoid arthritis due to its role in leukocyte recruitment. CD14 has connections to sepsis as it participates in the recognition of bacterial LPS leading to systemic inflammation. CD16 contributes to autoimmune diseases; its dysfunction can lead to impaired ADCC. CD68 levels have been studied in atherosclerosis linking it to foam cell formation and plaque stability. CD163 with its anti-inflammatory properties relates to inflammatory disorders like multiple sclerosis linked to hemoglobin-haptoglobin clearance and IL-10 production. These markers offer potential therapeutic targets for treating these diseases.

    Unpurified Anti-CD11b antibody [EP1345Y] - C-terminal ab52478 staining CD11b in the THP-1 (Human monocytic leukemia cell line) cell line by ICC/IF (Immunocytochemistry/immunofluorescence).

    Cells were fixed with 100% methanol. Samples were incubated with primary antibody (1/250). Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) ab150077 was used as the secondary antibody (1/1000). Nuclei were stained with DAPI.
