3D Tissue Staining Kit – CUBIC (ab316248) enables the easy set-up of 3D tissue imaging using simple protocols and standard laboratory equipment.
Together with the Tissue Clearing Kit – CUBIC (Tissue Clearing Kit – CUBIC ab316246) we provide a versatile tissue clearing and 3D tissue staining method that allows the observation of all cells in organs and the body. The kits are designed for safe and easy use in the lab.
The provided protocol solves issues pertaining to safety, waste processing, microscope compatibility, fluorescent protein signal retainability, etc.
To help you get started with tissue clearing, please visit the available data on tested antibodies on this datasheet. Validated antibodies are for key markers and are tested for tissue clearing and staining with whole mouse brains. If you wish to set up tissue clearing with antibodies that we haven’t tested yet, please consult the protocol book for further details.

After clearing the whole mouse brain with the Tissue clearing kit – CUBIC (Tissue Clearing Kit – CUBIC ab316246), anti-c-FOS Anti-c-Fos antibody [EPR21930-238] ab222699 was used with the 3D Tissue Staining Kit – CUBIC (ab316248).
White: nuclear staining, Yellow: c-FOS.
The sample was imaged using a light-sheet microscope.
Iba1 immunohistochemistry staining of whole mouse brain using rabbit anti-Iba1 antibody
Anti-Iba1 Anti-Iba1 antibody [EPR16588] ab178846 was used with Tissue clearing kit – CUBIC (Tissue Clearing Kit – CUBIC ab316246) and 3D Tissue Staining Kit – CUBIC (ab316248) to penetrate stain and clear a whole mouse brain.
Learn more about tissue clearing kits reagents and protocols designed to make it easier to stain whole brains and get more data from each valuable tissue sample.
For a whole mouse brain we recommend starting with 3.5 ug of Anti-Iba1 antibody [EPR16588] ab178846 and using a Fab fragment secondary antibody with 2.34 µg to create an antibody complex before 3D staining (see protocol for details). Additive A was used during the staining process.

NeuN immunohistochemistry staining of whole mouse brain using rabbit anti-NeuN antibody
Anti-NeuN Anti-NeuN antibody [EPR12763] - Neuronal Marker ab177487 was used with Tissue clearing kit – CUBIC (Tissue Clearing Kit – CUBIC ab316246) and 3D Tissue Staining Kit – CUBIC (ab316248) to penetrate stain and clear a whole mouse brain.
White: nuclear staining
Red: NeuN.

For a whole mouse brain we recommend starting with 10 ug of Anti-NeuN antibody [EPR12763] - Neuronal Marker ab177487 and using a Fab fragment secondary antibody with 6.67 µg to create an antibody complex before 3D staining (see protocol for details). Additive A was used during the staining process.
