The Albumin Creatinine Ratio Assay Kit (ab241018) provides a simple, sensitive, and high throughput adaptable assay that detects albumin (detection range: 0.02- 2.5 mg/mL), creatinine (detection range: 0.002 -0.5 mg/mL) and Albumin-to-creatinine ratio.

The ACR ratio is determined in two steps: First, albumin is determined by using a probe (Albumin Blue 580) that specifically recognizes albumin (Ex/Em = 600/630 nm). Second, creatinine is converted to sarcosine via enzymatic reactions. Arcosine is specifically oxidized generating a product that reacts with a probe producing a chromophore that can be detected at 570 nm.

Albumin/Creatinine often abbreviated as the albumin to creatinine ratio (ACR) serves as an important marker in the body. Albumin is a plasma protein with a molecular weight of approximately 66.5 kDa. It is synthesized in the liver and widely expressed in the blood while creatinine a breakdown product of creatine phosphate is a small molecule produced in the muscles and excreted in urine. The albumin/creatinine ratio measures these levels in urine to assess excretion balance and renal function.
Biological function summary

The albumin and creatinine levels help evaluate kidney function and filtration efficiency. Albumin signifies the presence of larger proteins in urine when normal kidney filtering mechanisms are impaired. The presence and concentration of creatinine meanwhile reflect waste removal efficacy. The albumin/creatinine ratio provides a reliable estimation of microalbuminuria which is an elevation in urinary albumin considered indicative of early kidney damage. This ratio is not directly part of a complex but offers insight into the normal physiological role of kidneys.

Albumin/creatinine measurements relate primarily to renal regulation and excretory pathways. They indicate how well glomeruli the filtering units in kidneys function relative to proteins and byproducts' reabsorption or excretion. Major calcium and magnesium transporter in plasma, binds approximately 45% of circulating calcium and magnesium in plasma (By similarity). Potentially has more than two calcium-binding sites and might additionally bind calcium in a non-specific manner (By similarity). The shared binding site between zinc and calcium at residue Asp-273 suggests a crosstalk between zinc and calcium transport in the blood (By similarity). The rank order of affinity is zinc > calcium > magnesium (By similarity). Binds to the bacterial siderophore enterobactin and inhibits enterobactin-mediated iron uptake of E.coli from ferric transferrin, and may thereby limit the utilization of iron and growth of enteric bacteria such as E.coli (PubMed : 6234017).
