Catalyzes the NADPH-dependent reduction of a wide variety of carbonyl-containing compounds to their corresponding alcohols. Displays enzymatic activity towards endogenous metabolites such as aromatic and aliphatic aldehydes, ketones, monosaccharides and bile acids, with a preference for negatively charged substrates, such as glucuronate and succinic semialdehyde (PubMed:10510318). Functions as a detoxifiying enzyme by reducing a range of toxic aldehydes. Reduces methylglyoxal and 3-deoxyglucosone, which are present at elevated levels under hyperglycemic conditions and are cytotoxic. Involved also in the detoxification of lipid-derived aldehydes like acrolein (By similarity). Plays a role in the activation of procarcinogens, such as polycyclic aromatic hydrocarbon trans-dihydrodiols, and in the metabolism of various xenobiotics and drugs, including the anthracyclines doxorubicin (DOX) and daunorubicin (DAUN) (PubMed:18276838, PubMed:11306097). Aldo-Keto Reductase (AKR) Activity Assay Kit (Colorimetric) (ab211112) provides a convenient tool for sensitive detection of Aldo-Keto Reductase (AKR) activity in animal tissue and cells, as well as biological fluids such as serum. The assay is based on the ability of AKR to reduce a general substrate and convert NADP+ to NADPH, which reacts with the AKR probe to generates color that can be detected at OD 450 nm. The intensity of the color is proportional to the activity of AKR in the sample.
This assay can detect as low as 1 μU of AKR activity in samples. It has been validated with AKR1B10, AKR1C1 and AKR1C3.
This product is manufactured by BioVision, an Abcam company and was previously called K847 EZDetect™ Aldo-keto Reductase Activity Assay Kit (Colorimetric). K847-100 is the same size as the 100 test size of ab211112.
Aldo-keto Reductases (AKRs) are a superfamily of NAD(P)H linked oxidoreductases which contains more than 190 members and are present in nearly all phyla. They are mainly monomeric soluble proteins (34 – 37 kD) and their common is to oxidize/reduce aldehydes or ketones to their corresponding primary or secondary alcohols. Due to their broad substrate specificity, AKRs play roles in intermediary metabolism, biosynthesis and detoxification.
In humans, AKRs are not only involved in carbonyl metabolism, but they also regulate signaling through nuclear receptors which leads to chemical carcinogenesis. Many studies demonstrate increasing expression of AKRs in cancers such as lung, liver and colon cancers.

AKR1A1 also known as aldehyde reductase or aldo-keto reductase is a member of the aldo-keto reductase family. It functions mechanically as a reductase enzyme that converts aldehydes and ketones to their corresponding alcohols via NADPH as a cofactor. The enzyme has a molecular mass of approximately 36 kDa. AKR1A1 is expressed in a wide range of tissues including liver kidney and brain indicating its broad role in metabolic processes.
AKR1A1 is involved in the detoxification of aldehydes produced during metabolism and oxidative stress. This function occurs as part of a larger detoxification system. Being part of the aldo-keto reductase family it contributes to maintaining homeostasis by regulating the levels of reactive aldehydes. Through this activity it affects cellular responses to stress and the metabolism of xenobiotics.
AKR1A1 has significant involvement in metabolic and detoxification pathways notably in glycolysis and xenobiotic metabolism. It interacts with various aldehydes formed as intermediates in these pathways. Related proteins in these pathways include AKR1B1 which also plays a role in reducing sugar-derived aldehydes emphasizing the importance of AKR1A1 in controlling toxic aldehyde levels and protecting cellular components from damage.
AKR1A1 has been linked to diabetic complications due to its role in polyol pathway where it contributes to the conversion of glucose to sorbitol. This pathway can lead to osmotic and oxidative stress when dysregulated contributing to retinal and renal dysfunctions in diabetes. Additionally AKR1A1 is connected to cancer development where it affects oxidative stress levels and tumor progression alongside proteins like AKR1C1 which are also involved in lipid and steroid metabolism further influencing disease outcomes.
