Alpha-Amylase enzyme is responsible for the breakdown of complex carbohydrates in humans. Thus, inhibition of α-amylase could be considered as a strategy for the treatment of disorders related to carbohydrate uptake, such as diabetes, obesity, dental cavities and periodontal diseases. In this kit human Alpha-Amylase hydrolyzes the synthetic substrate, yielding smaller fragments containing the chromophore (pNP; OD = 405 nm). A potent, specific inhibitor is also included in the kit.
The 1-step assay is simple and can be completed within 1 hour.
This product is manufactured by BioVision, an Abcam company and was previously called K482 a-Amylase Inhibitor Screening Kit. K482-100 is the same size as the 100 test size of ab283391.

Pancreatic alpha amylase also known as alpha-amylase P or AMY2A and salivary alpha amylase (AMY1) are enzymes that play essential roles in carbohydrate digestion. Mechanically they hydrolyze alpha-14-glycosidic linkages in starches breaking them down into smaller sugars like maltose and dextrins. Pancreatic alpha amylase has a molecular mass of approximately 56 kDa. The pancreas and salivary glands predominantly express these enzymes facilitating digestive processes in the small intestine and oral cavity respectively.
These alpha-amylases initiate the digestive process for dietary starches and glycogen by breaking them into sugars which are then more easily absorbed by the body. While they function largely independently their activity is supported by other digestive enzymes that further process carbohydrates throughout the digestive system. This enzymatic action is important for maintaining energy supply from glycogen and starches highlighting the significant role of alpha amylase in metabolic nutrition.
The catalytic action of pancreatic and salivary alpha amylase is a part of the digestive pathway for carbohydrates where it accelerates the breakdown of starches. These enzymes interact within the amylase pathway which includes other related enzymes like alpha-glucosidases that sequentially digest disaccharides into glucose. They are not known to form permanent complexes but work in conjunction with these digestive enzymes to facilitate efficient energy release.
Alterations in alpha-amylase activity have connections to conditions like pancreatitis and salivary gland dysfunctions where changes in enzyme levels provide important diagnostic information. Conditions such as pancreatitis an inflammation of the pancreas link directly to abnormal amylase activity and concentration.
