Beta Lactamase Activity Assay Kit (Colorimetric) ab197008 is a simple and sensitive assay that can detect and quantify the enzymatic activity of beta lactamases, a large family of hydrolases.
The beta lactamase assay protocol is based on the hydrolysis of the substrate Nitrocefin, a chromogenic cephalosporin, that results in the generation of a colored product (detectable at OD=490 nm), which is directly proportional to the amount of beta-lactamase activity.
This product can detect as low as 0.06 mU of beta-lactamase activity from bacterial samples or from biological fluids infected with beta-lactamase expressing bacteria.

This product is manufactured by BioVision, an Abcam company and was previously called K803 Beta-Lactamase Activity Colorimetric Assay Kit. K803-100 is the same size as the 100 test size of ab197008.
Beta Lactamases (β-Lactamases, βLs), are a large family of hydrolases comprising more than 850 identified members expressed in Gram-positive and Gram-negative bacteria. βLs can be classified according to their substrate or inhibitor specificity. These enzymes are capable of hydrolyzing four atom rings known as β-lactams.

Beta lactamase sometimes referred to as penicillinase is an enzyme with a mass typically around 29 kDa. This enzyme hydrolyzes the beta-lactam ring an important structure in beta-lactam antibiotics which make this antibiotics ineffective. Beta lactamase is expressed in diverse bacterial species where its presence is a marker of antibiotic resistance. Scientists have developed inhibitors to block its action which are used in combination with antibiotics to combat resistant bacterial strains.
Beta lactamase facilitates the survival of bacteria in the presence of beta-lactam antibiotics by breaking down these drugs. It is often part of a larger complex that includes various resistant genes enhancing its capacity to resist antibiotic pressures. The enzyme helps bacterial cells bypass the effects of these antibiotics by deactivating them before they can inhibit cell wall synthesis.
Beta lactamase operates within the bacterial resistance mechanism directly impacting the beta-lactam antibiotic degradation pathway. This enzyme works alongside other resistance proteins like penicillin-binding proteins (PBPs) which are involved in cell wall biosynthesis. Their combined actions enable pathogenic bacteria to persist in environments with high antibiotic concentrations complicating disease treatment.
Beta lactamase has significant impact on bacterial infections such as those caused by Staphylococcus aureus and Escherichia coli which can lead to disorders like pneumonia and urinary tract infections. Its activity is linked to the failure of treatment with beta-lactam antibiotics. Related resistance proteins like metallo-beta-lactamases extend this resistance to even broader antibiotic classes presenting further challenges in managing these infections.
