Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - BOND RX - Anti-CD73 antibody [EPR6114] (AB321953)

Immunohistochemical analysis of paraffin-embedded human liver tissue labelling CD73 with ab133582 [EPR6114] at 2.2 µg/mL concentration.

All steps were performed at room temperature unless otherwise stated.

Staining was performed on the BOND RX Advanced Staining Instrument from Leica Biosystems, using the BOND Polymer Refine Detection kit (DS9800) and the following protocols :

  1. Deparaffinization - Dewax
  2. Antigen retrieval - HIER with ER2 (pH 9.0), 20 minutes, 100°C
  3. Staining – modified IHC Protocol F (Marker : ab133582 for 15 minutes with 3 x BOND Wash Solution steps) where standard post-primary step has been excluded.

Sections were counterstained with Hematoxylin (DS9800).

Secondary Antibody only control was performed with the same staining protocols, with marker as negative.

This product is a bundle of 2 x BOND Titration Containers (with 6mL inserts) along with 100 uL vial of Anti-CD73 antibody [EPR6114] - ab133582.

Additional Titration Inserts can be purchased from Leica Biosystems, using OPT9719 (box of 50). Or Titration Container/Insert Kit, using OPT9049 (box of 10/50).

CD73 also known as ecto-5'-nucleotidase is an enzyme that functions at the cellular level. The CD73 protein hydrolyzes extracellular nucleotides into nucleosides by removing phosphate groups. It is a 70 kDa glycosylphosphatidylinositol-anchored molecule found on the surface of various cell types including endothelial cells lymphocytes and epithelial cells. Its presence is noted in a wide range of tissues across the human body highlighting its versatile role in cellular communication and metabolism.
Biological function summary

CD73 participates in the regulation of purinergic signaling through adenosine production. It acts in cooperation with other cell surface enzymes such as CD39 forming a functional complex that handles ATP breakdown to adenosine. This enzymatic activity modulates immune responses tissue protection and inflammation control. In addition to immune-related functions CD73 supports the maintenance of vascular integrity and proper cellular adhesion which is important in different physiological and pathological contexts.

CD73 plays an important part in the adenosinergic pathway governing the conversion of AMP into adenosine.
