Marker Expression : Cortical Glutamatergic Neurons (ab315366) have high neuronal purity (>90%) and comprise predominantly excitatory neurons. Labeling with the pan-neuronal marker MAP2 (red) highlights the neuronal purity of these cells. Labeling with the glutamatergic-specific markers CTIP2 (green) and TBR1 (blue) indicates overwhelming presence of glutamatergic neurons.

For even greater physiological relevance, co-culture protocols are available for use with Cortical astrocytes (ab315369). Co-culture with Cortical astrocytes can be performed with Cortical Glutamatergic Neurons, Cortical GABAergic Neurons (ab315368) or a combination of both in varying ratios.

A GFP-expressing alternative to this product is also available (ab315367).

Human Cortical Glutamatergic Neurons are derived from normal control iPSC line WC-30. BrainXell Cortical Glutamatergic Neurons have high neuronal purity (>80%) and comprise predominantly excitatory neurons. Labelling with the pan-neuronal marker MAP2 highlights the neuronal purity of these cells. Labelling with the glutamatergic-specific marker FOXP2 indicates overwhelming presence of glutamatergic neurons. Cortical Glutamatergic Neurons are adherent and exhibit substantial neurite outgrowth within the first week in culture, showing the characteristic oval cell shape and long processes of classic cortical glutamatergic neurons. These neurons also demonstrate pronounced electrophysiological activity after two weeks in culture, as demonstrated by multi-electrode array (MEA) recordings.

Vials of Cortical Glutamatergic Neurons are delivered with three supplements which are designed to facilitate the maturation and health of the cultures. BrainFast Glut is a seeding supplement which promotes cell survival at thaw and completes the differentiation process from late-stage neural progenitor to neuron. BrainFast SK enhances neuronal purity, and BrainFast D4 accelerates maturation.
