ab325358 Factor Xa Activity Assay Kit (Fluorimetric) utilizes the ability of Factor Xa to cleave a synthetic peptide substrate and release AMC (fluorophore), which can be easily quantified by fluorescence at Ex/Em = 368/460 nm. The fluorometric signal is directly proportional to the Factor Xa activity in samples. Our assay kit can detect as low as 1 ng, or 35 µU, of FXa activity in biological samples.

Product details

Factor Xa (FXa) is the activated form of the coagulation factor X (Stuart-Power factor, thrombokinase, prothrombinase, thromboplastin, E.C.3.4.21.6). Factor X, a serine endopeptidase plays an important role at several stages of the coagulation pathway. It acts by converting prothrombin into active thrombin by complexing with activated co-factor V in the prothrombinase complex. Unfractionated heparin and various low molecular weight heparins bind to plasma cofactor antithrombin to inactivate several coagulation factors including factor Xa. Abcam's Factor Xa activity assay kit utilizes the ability of Factor Xa to cleave a synthetic peptide substrate and release AMC (fluorophore), which can be easily quantified by fluorescence at Ex/Em = 368/460 nm. The fluorometric signal is directly proportional to the Factor Xa activity in samples. Our assay kit can detect as low as 1 ng, or 35 µU, of FXa activity in biological samples.

Heparin-treated plasma is not recommended for the assay. This is because Heparin binds to plasma cofactor antithrombin to inactivate several coagulation factors, including Factor Xa.

•    Thaw kit components and aliquot components susceptible to freeze/thaws; get equipment ready.
•    Prepare standard curve and positive control reactions.
•    Prepare samples in duplicate (find optimal dilutions to fit standard curve readings).
•    Set up plate for standard, positive control, and samples (50 µL).
•    Prepare FXa Reaction Mix.

Factor Xa is a vitamin K-dependent glycoprotein that converts prothrombin to thrombin in the presence of factor Va, calcium and phospholipid during blood clotting (PubMed : 22409427). Factor Xa activates pro-inflammatory signaling pathways in a protease-activated receptor (PAR)-dependent manner (PubMed : 24041930, PubMed : 30568593, PubMed : 34831181, PubMed : 18202198). Up-regulates expression of protease-activated receptors (PARs) F2R, F2RL1 and F2RL2 in dermal microvascular endothelial cells (PubMed : 35738824). Triggers the production of pro-inflammatory cytokines, such as MCP-1/CCL2 and IL6, in cardiac fibroblasts and umbilical vein endothelial cells in PAR-1/F2R-dependent manner (PubMed : 30568593, PubMed : 34831181). Triggers the production of pro-inflammatory cytokines, such as MCP-1/CCL2, IL6, TNF-alpha/TNF, IL-1beta/IL1B, IL8/CXCL8 and IL18, in endothelial cells and atrial tissues (PubMed : 24041930, PubMed : 35738824, PubMed : 9780208).
