Factor XIa also known as Coagulation Factor XIa or FXIa plays an important role in the blood coagulation cascade. This serine protease has a molecular weight of approximately 160 kDa and derives from the zymogen Factor XI. Factor XIa is synthesized in the liver and circulates in the blood plasma. In its active form it cleaves the Arg369-Ile370 bond in Factor IX leading to the further activation of the coagulation pathway.
Factor XIa is essential for hemostasis. It forms a part of the intrinsic pathway complex that contributes to the amplification of thrombin generation. This activity strengthens the formation of a stable fibrin clot which is critical to prevent excessive bleeding. Factor XIa is also involved in cellular response mechanisms beyond coagulation such as inflammatory processes.
Factor XIa is a significant component of the intrinsic coagulation pathway and the contact activation system. It works closely with proteins like Factor IX and kallikrein facilitating the transformation of inactive proteins to their active states in a stepwise manner. These interactions support hemostatic balance and help maintain proper clot formation while preventing thrombosis.
Factor XIa has connections to thrombotic conditions such as deep vein thrombosis and ischemic stroke. Excessive activation of FXIa can contribute to pathological clot formation. Inhibitors like anti-XIa reduce activity and are explored for therapeutic interventions in these conditions. Additionally congenital Factor XI deficiency often called Hemophilia C presents a bleeding disorder that highlights FXIa's significance in coagulation.
