Abcam's Glucose-6-Phosphate Assay Kit - High Sensitivity (Fluorometric) (ab107923) is a simple, sensitive and rapid means of quantifying glucose-6-Phosphate (G6P) in a variety of samples. In the assay, glucose-6-phosphate is oxidized with the generation of a product that converts a nearly colorless probe to an intensely fluorescent product (Ex/Em 535/587 nm). This assay can detect G6P in the range of 10 to 500 pmoles which is equivalent to the range of 1-500 μM in the original sample assuming a dilution of 5X during processing. This Glucose-6-Phosphate Assay Kit - High Sensitivity is more sensitive than our colorimetric assay, Glucose-6-Phosphate Assay Kit (ab83426).

This product is manufactured by BioVision, an Abcam company and was previously called K687 PicoProbe™ Glucose-6-Phosphate Fluorometric Assay Kit. K687-100 is the same size as the 100 test size of ab107923.

Glucose-6-phosphate (G6P) is a key intermediate for glucose transport into cells which then enters either metabolic pathways or storage. G6P can enter the glycolytic pathway, the pentose phosphate shunt or be stored as glycogen or starch. G6P is utilized by its dehydrogenase to generate reducing equivalents in the form of NAD(P)H. This is particularly important in red blood cells where a G6PDH deficiency leads to hemolytic anemia.

Glucose 6-phosphate also known as G6P is an essential intermediary metabolite in cellular functions. It serves as a pivotal molecule in various metabolic pathways. Glucose 6-phosphate forms during the first step of glycolysis through the phosphorylation of glucose by the enzyme hexokinase a reaction that adds a phosphate group. The molecular mass of glucose 6-phosphate is approximately 260.14 g/mol. It is expressed in most cell types throughout the body playing a central role in energy metabolism.
Biological function summary

Glucose 6-phosphate acts as a substrate for several biochemical processes within the cell. It is an important component in glycogen synthesis and the pentose phosphate pathway providing important intermediates for nucleic acid and lipid synthesis. Although it is not part of a complex in itself glucose 6-phosphate interacts with various enzymes to activate these pathways reflecting its functional significance.

Pathways

Glucose 6-phosphate is a critical connector in glycolysis and gluconeogenesis. In glycolysis the molecule continues through a series of enzymatic reactions to generate ATP which cells use for energy. Meanwhile in gluconeogenesis glucose 6-phosphate can be converted back to glucose. G6P acts in coordination with enzymes such as glucose-6-phosphate dehydrogenase in the pentose phosphate pathway highlighting its multifunctional nature and its interaction with other proteins like phosphoglucomutase.

Glucose 6-phosphate has significant implications in diabetes and glycogen storage diseases. Diabetes characterized by impaired glucose regulation involves altered levels of glucose 6-phosphate. Aberrant function of glucose 6-phosphatase an enzyme that converts glucose 6-phosphate to glucose can lead to glycogen storage diseases like von Gierke's disease. These disorders connect with glucose 6-phosphate through the disrupted metabolic process underlining its critical role in maintaining normal glucose homeostasis.
