Product details

Glucose Oxidase Assay Kit ab138884 uses a quick and sensitive fluorometric assay for the measurement of glucose oxidase activity. It can be performed in a 96-well or 384-well microtiter plate format and is readily adapted to automation without a separation step.

The glucose oxidase assay protocol uses a red dye which enables a dual recordable mode. The fluorescent signal can be easily read by either a fluorescence microplate reader at Ex/Em = 540/590 nm or an absorbance microplate reader at 576 nm.

Glucose oxidase assay protocol summary:
- add standards and samples to wells
- add reaction mix to wells and incubate for 10-30 min at 37°C
- analyze with a microplate reader

Glucose oxidase is a dimeric protein that catalyzes the oxidation of beta-D-glucose into hydrogen peroxide and D-glucono-1,5-lactone, which is hydrolyzed to gluconic acid. It is widely used for the determination of glucose in body fluids and in removing residual glucose and oxygen from beverages, food and other agricultural products. Furthermore, Glucose oxidase is commonly used in biosensors to detect glucose.

Supplementary information

Glucose oxidase also known as GOX is an enzyme that catalyzes the oxidation of beta-D-glucose to glucono delta-lactone releasing hydrogen peroxide as a by-product. This enzyme with a molecular weight of approximately 160 kDa is commonly found in fungi like Aspergillus and Penicillium species. Glucose oxidase is predominantly expressed in the extracellular space of these organisms facilitating its role in glucose metabolism and defense mechanisms. It plays an important role in various biochemical assays including the 'glucose oxidase assay' 'glucose oxidase test' and 'glucose oxidase kit' often used for glucose quantification in diverse samples.
Biological function summary

Glucose oxidase is significant in both metabolic pathways and environmental defense. The enzyme acts as a part of a larger enzymatic reaction complex that includes peroxidase which together help in the glucose oxidase reaction to maintain cellular redox balance. Additionally the production of hydrogen peroxide by glucose oxidase serves as a chemical defense mechanism against microbial infections. This property is harnessed in technical applications making glucose oxidase a valuable tool in biotechnology for both its metabolic impacts and antimicrobial properties.

Pathways

Glucose oxidase is an important enzyme in the carbohydrate metabolism and oxidative stress response pathways. It acts by converting glucose into glucono delta-lactone which can then enter further metabolic processes. The enzyme interacts closely with other proteins particularly those involved in oxidative phosphorylation such as catalase which neutralizes the hydrogen peroxide generated during the glucose oxidase reaction. These interactions help modulate cellular energy production and maintain oxidative homeostasis highlighting its role within broader metabolic networks.

Glucose oxidase activity is relevant to conditions such as diabetes and neurodegenerative diseases. The enzyme's function in glucose monitoring tools plays a critical role in diabetes management where accurate glucose measurement is important. Additionally altered levels of hydrogen peroxide due to dysregulated glucose oxidase activity is implicated in oxidative stress which can contribute to neurodegenerative disorders like Alzheimer's disease. In these contexts the enzyme's interaction with proteins like catalase and superoxide dismutase becomes significant as these proteins aid in mitigating oxidative damage which is central to the pathology of such diseases.

Product protocols

Target data

Glucose oxidase catalyzes the oxidation of beta-D-glucose to D-glucono-delta-lactone and hydrogen peroxide in the presence of molecular oxygen. D-glucono-delta-lactone is sequentially hydrolyzed by lactonase to D-gluconic acid, and the resulting hydrogen peroxide is hydrolyzed by catalase to oxygen and water (PubMed : 14188176, PubMed : 14257628, PubMed : 14299649, PubMed : 15450808, PubMed : 2076553, PubMed : 2406261, PubMed : 24283586, PubMed : 28631058, PubMed : 28939970, PubMed : 34500289, PubMed : 7826581, Ref.4, Ref.5). The activity shows high specificity to beta-D-glucose, with very low to no activity towards L-glucose, 2-deoxy-D-glucose, 3-deoxy-D-glucose, 4-deoxy-D-glucose, 5-deoxy-D-glucose, 6-deoxy-D-glucose, 3-O-methyl-D-glucose, 4-O-methyl-D-glucose, 6-O-methyl-D-glucose, 4,6-O-benzylidene-D-glucose, 5-thio-5-deoxy-D-glucose, D-mannose, D-allose, D-galactose, D-fructose, D-arabinose, D-xylose, trehalose, melibiose, L-mannomethylose, lactose, sucrose or 1,5-anhydro-D-glucitol (PubMed : 14188176, PubMed : 14257628, PubMed : 24283586, Ref.5).
