Beta-Glucuronidase Activity Assay Kit (Fluorometric) (ab234625) provides a quick, reliable fluorometric method for measurements of Beta-Glucuronidase activities of samples and tissue lysates. The provided substrate, which is specific to Beta-Glucuronidases, is cleaved into a fluorescent product in the presence of Beta-Glucuronidase. This kit, when used according to the protocol, is sensitive enough to detect as little as one uU (1 pmol/min) of activity.

Beta-Glucuronidases are hydrolytic enzymes responsible for the breakdown of carbohydrates. Specifically, Beta-Glucuronidases cleave the terminal Beta-D-glucuronic acid residue from the non-reducing terminus of a mucopolysaccharide chain. In humans, these enzymes are found in the lysosome of many tissue types. Loss of Beta-Glucuronidase activity results in a metabolic disease known as Sly syndrome. One pharmaceutical application for these enzymes is the metabolism of glucuronidated prodrugs into active pharmacological compounds. As expression and activities of Beta-Glucuronidases vary substantially between tissue types and disease states, these enzymes have been used to achieve targeted activation of oncotherapeutic compounds, some of which may be toxic to healthy cells not associated with malignancy or disease. It is thus important to have knowledge of the Beta-Glucuronidase activity in the tested sample to determine whether the prodrug or active form will predominate.

Beta-glucuronidase sometimes called β-glucuronidase or b-glucuronidase is an enzyme that catalyzes the hydrolysis of beta-glucuronides into aglycons and D-glucuronic acid. It exhibits a molecular mass of approximately 290 to 332 kDa depending on its source and post-translational modifications. This enzyme is expressed in lysosomes of various tissues including the liver spleen and kidneys. Its activities are important in processes like drug metabolism and the breakdown of complex carbohydrates.
Biological function summary

Beta-glucuronidase functions play a role in the cleavage of glycosaminoglycans which are important structural components of cellular and extracellular environments. It works mainly as a monomer but in some cases can form complexes with other proteins to stabilize its function. By breaking down glucuronic acid-containing polysaccharides beta-glucuronidase helps recycle and reuse cellular carbohydrates which are key for maintaining the cellular matrix and intercellular communication.

Pathways

Beta-glucuronidase activity is central to the lysosomal degradation pathway and has significant implications in the hormone regulation pathway. The enzyme interacts with other proteins involved in lysosomal storage and transport contributing to the catabolism of glycosaminoglycans and modulation of active hormone levels. Beta-glucuronidase works closely with arylsulfatase A in the degrading pathway ensuring efficient recycling of sulfated glycosaminoglycans and steroid hormones.

Disruptions in beta-glucuronidase activity have connections to serious health issues such as mucopolysaccharidosis type VII and certain forms of cancer. In mucopolysaccharidosis type VII or Sly syndrome an inherited disorder lack of beta-glucuronidase leads to harmful accumulation of glycosaminoglycans. In cancer altered beta-glucuronidase levels can affect drug metabolism and hormone modulation influencing disease progression and treatment efficacy. Researchers also explore connections between beta-glucuronidase and other proteins such as hyaluronidase which together may influence cancer metastasis and the breakdown of extracellular matrix components.
