Nitrite concentration in pooled human serum and HeLa nuclear extracts.
Human serum was deproteinized using 10 kDa columns. Filtrate (50 μl, undiluted) was collected and assayed according to kit protocols. HeLa nuclear cells were homogenized using Nitrite Assay Buffer (100 μg protein).

Griess Assay Kit / Griess Reagent Kit ab234044 uses a classic protocol for the estimation of nitrite in biological samples.
Griess assay principle
In the Griess reaction, Nitrite is reduced to nitrogen oxide using Griess Reagent I. Nitrogen oxide then reacts with Griess Reagent II forming a stable product that can be detected by its absorbance at 540 nm.
Griess Reagent I is 1% Sulfanilamide in 5% Phosphoric Acid.
Griess Reagent II is 0.1% NED (N-(1-Naphthyl)ethylenediamine dihydrochloride) in water.
The two-step assay is simple, fast and can detect nitrite levels as low as 1 nmol/well.
Griess assay protocol summary
- add samples and standards to wells
- sequentially add Griess Reagent I, Griess Reagent II, assay buffer to wells
- incubate for 10-min at room temperature
- analyse on a plate reader
