Heme Assay Kit (ab272534) is a simple, direct and automation-ready procedure for measuring heme concentration. This Assay Kit is based on an improved aqueous alkaline solution method, in which the heme is converted into a uniform colored form. The intensity of color, measured at 400 nm, is directly proportional to the heme concentration in the sample. The optimized formulation substantially reduces interference by substances in the raw samples and exhibits high sensitivity.

Sensitive and accurate: Linear detection range 0.6-125 uM heme in 96-well plate assay.

Simple and high-throughput: The "mix-and-read" procedure involves addition of a single working reagent and reading the optical density. Can be readily automated as a high-throughput assay in 96-well plates for thousands of samples per day.

Safety: Reagents are non-toxic.

Versatility: Assays can be executed in 96-well plate or cuvette.

Heme often referred to as haem is a small molecule with a mass of approximately 616.5 g/mol. This iron-containing porphyrin is an essential component of various hemoproteins which include hemoglobin and myoglobin. Heme is expressed in several tissues predominantly within the liver bone marrow and muscles. It plays an important role in oxygen transportation and storage serving as the prosthetic group that enables hemoglobin to carry oxygen efficiently.
Biological function summary

Heme plays a critical role in several cellular processes beyond oxygen transport. It functions as a cofactor in enzymatic reactions involving enzymes like cytochrome P450 and catalase. As part of the cytochrome complex it aids in electron transfer during mitochondrial electron transport chain processes important for ATP production. The concentration of heme within cells is tightly regulated through synthesis and degradation pathways ensuring cellular functions maintain balance.

Pathways

Heme influences cellular functions particularly in the heme biosynthesis and degradation pathways.
