L-Amino Acid Assay Kit ab65347 is used measure the level of L-amino acids in biological samples. There is no requirement for sample pretreatment or purification.

The L-amino acid(s) level can be quantified using fluorometric (at Ex/Em = 535/587 nm) or colorimetric (570 nm) methods in 96-well plates.

L-amino acids are enantiomeric forms of amino acids which are the building blocks of proteins. These molecules exhibit chirality existing as either L- or D-isomers with L-amino acids being the form utilized in biological systems. L-amino acids are found in almost all proteins and enzymes across various living organisms from bacteria to humans. They contribute to the structural stability and function of proteins. These molecules have different molecular weights typically ranging from about 75 to 204 Da depending on the specific amino acid. L-amino acids are expressed ubiquitously in cellular proteins reflecting their fundamental role in life.
Biological function summary

L-amino acids serve as the basic units of proteins generated through the polymerization process during protein synthesis. This process involves the ribosomal complex where L-amino acids bind to transfer RNA (tRNA) molecules following the genetic instructions encoded in mRNA. L-amino acids are essential for the formation of the three-dimensional structure and functional specificity of proteins. Beyond protein synthesis L-amino acids play a role in metabolic pathways being precursors for hormones neurotransmitters and other bioactive molecules.

Pathways

L-amino acids participate in various biochemical routes such as the citric acid cycle and nitrogen metabolism. They enter the citric acid cycle primarily through their deamination contributing to the production of energy by providing intermediates such as alpha-ketoglutarate. In nitrogen metabolism L-amino acids serve as donors of amino groups an important step for the synthesis of nucleotides and other nitrogen-containing compounds. They interact closely with enzymes like aminotransferases in these pathways highlighting their versatility in metabolic functions.

L-amino acids connect to conditions such as phenylketonuria (PKU) and maple syrup urine disease (MSUD). PKU results from a defect in phenylalanine metabolism leading to the accumulation of this L-amino acid and resulting in potential neurological damage if untreated. In MSUD the body's inability to break down certain branched-chain L-amino acids causes these acids to accumulate which can result in severe neurological issues and metabolic crises. Disturbances in these metabolic pathways and enzyme deficiencies illustrate the critical role of L-amino acids and their related metabolic enzymes in maintaining health.
