Supplementary information

Hepatitis B virus (HBV) is a small DNA virus with a mass of about 3.2 kilobases known to cause liver infection. It primarily targets hepatocytes the main cells of the liver resulting in inflammation and damage. Orcein staining a technique known for highlighting elastic fiber structures in tissue samples occasionally detects HBV as orcein dye binds to the virus-infected cells highlighting elastic fibers in histology studies. Orcein-derived stains are also known to be useful in the histological analysis of liver biopsies when diagnosing HBV infection. This virus is also referred to as serum hepatitis given it is transmitted through exposure to infectious body fluids such as blood.
Biological function summary

The hepatitis B virus affects liver cell function by integrating into the host genome influencing liver metabolism and immune responses. It forms a complex with the hepatitis B surface antigen (HBsAg) that facilitates viral assembly and secretion. The orcein stain histology method helps reveal changes in liver architecture as a result of ongoing HBV replication illustrating the impact on elastic fibers within tissues. Elastic fiber staining such as with orcein further assists in understanding the tissue-level changes in chronic hepatitis patients.

Pathways

Hepatitis B virus influences several cellular processes including the PI3K/Akt pathway. This pathway affects cell survival proliferation and metabolism often associated with hepatitis B surface antigen. HBV can modulate the NF-kB pathway which is important for immune responses and inflammation regulation. Through these pathways HBV affects interactions with other proteins such as Akt and NF-kB important for maintaining cell homeostasis and defense mechanisms against viral infections.

Hepatitis B virus is a leading cause of chronic hepatitis and liver cancer (hepatocellular carcinoma). Chronic infection with HBV elevates inflammation and fibrosis often detected through histological methods like orcein staining. This connection emphasizes the relationship between HBV and altered extracellular matrix components including elastic fibers. Additionally proteins such as p53 involved in cell cycle regulation and apoptosis may interact with HBV during carcinogenic processes highlighting its role in liver disease progression.
