Product details

The Ornithine Assay Kit (Fluorometric) (ab252903) provides a rapid, specific, and easy method for the measurement of total ornithine concentrations in a wide variety of biological samples. In this enzyme-based assay, ornithine is converted into a series of intermediates, which will further react with a probe producing a stable fluorometric signal (Ex/Em = 535/587nm). The kit is simple to use, sensitive and high-throughput adaptable and can detect as low as 50 pmol/well of ornithine in biological samples.

This product is manufactured by BioVision, an Abcam company and was previously called K939 Ornithine Assay Kit (Fluorometric). K939-100 is the same size as the 100 test size of ab252903.

Supplementary information

Ornithine sometimes referred to as L-ornithine is an amino acid without protein-coding properties acting primarily in the urea cycle. It has a mass of approximately 132.16 g/mol. Unlike many other amino acids ornithine does not incorporate into proteins but plays a critical role in cellular metabolism. In terms of expression ornithine commonly remains within liver cells where it contributes to the detoxification process of ammonia a byproduct of digestion and cellular respiration.
Biological function summary

Ornithine operates as an integral part of the urea cycle facilitating the conversion of ammonia into urea. Within this cycle it serves as a substrate for the enzyme ornithine transcarbamylase leading to the production of citrulline. This role situates ornithine within a complex cycle important for nitrogen disposal. Beyond the liver ornithine levels may impact tissues linked to high protein turnover as it interfaces with other amino acids like arginine and citrulline in protein metabolism.

Pathways

Ornithine figures prominently in two main pathways: the urea cycle and the polyamine biosynthesis pathway. Within the urea cycle it closely relates to enzymes like carbamoyl phosphate synthetase I and ornithine transcarbamylase. This cycle interfaces with pathways responsible for amino acid breakdown. In the polyamine biosynthesis pathway ornithine converts into putrescine a precursor for polyamines such as spermidine and spermine linking to pathways that regulate cell growth and differentiation.

Ornithine’s imbalance can correlate with some metabolic disorders like hyperammonemia and ornithine transcarbamylase deficiency where the urea cycle malfunction results in excessive ammonia levels. This deficiency often involves mutations in the ornithine transcarbamylase enzyme. Alterations in the polyamine biosynthesis pathway involving overproduction of polyamines also link to disorders such as cancer. Connections between ornithine enzymes of the urea cycle and polyamine metabolism indicate its importance in both normal physiological processes and disease states.
