Quantitation of OxLDL in Serum and Plasma Samples.

Left : LDL Recovery After Precipitation Solution. Serum and plasma samples were treated with LDL Precipitation Solution according to the Sample Preparation protocol. LDL recovery was determined. Right : OxLDL Determination of Serum and Plasma Samples. Serum and plasma samples were treated with LDL Precipitation Solution according to the Sample Preparation Section. Precipitated LDL pellets were resuspended in 1.6 mL of PBS before further diluting 1 : 50 in Assay Diluent. Samples were tested according to the Assay Protocol.

The Oxidized LDL Assay Kit (HNE-LDL, Human) ELISA Kit (ab242304) is developed for the detection and quantitation of human oxLDL in plasma, serum or other biological fluid samples.

The kit contains a copper oxidized LDL standard and has a detection sensitivity limit of ~150 ng/mL. Each kit provides sufficient reagents to perform up to 96 assays including standard curve and unknown samples.

Oxidized phospholipids often referred to as oxidized LDL or ox-LDL play a role in lipid metabolism and atherosclerosis development. Ox-LDL results from the oxidative modification of low-density lipoprotein (LDL) molecules heavily impacting cardiovascular health. The major component of ox-LDL malondialdehyde-modified LDL (MDA-LDL) expresses on the surface of endothelial cells macrophages and smooth muscle cells. This oxidative process increases the atherogenic properties of LDL contributing to its accumulation in arterial walls.
Biological function summary

Oxidized phospholipids strongly influence inflammation and immune responses. These molecules act within cell membranes triggering toll-like receptors (TLRs) and other immune sensors. Ox-LDL forms part of immunoglobulin complexes and interacts with scavenger receptors like CD36 on macrophages. It recruits immune cells and promotes the release of cytokines perpetuating an inflammatory environment within blood vessels.

Pathways

Oxidized phospholipids participate in lipid oxidative stress and inflammatory pathways. They contribute significantly to the pathophysiology of the atherosclerotic process. In the oxidative stress pathways ox-LDL affects the balance of reactive oxygen species (ROS) and nitric oxide (NO) levels influencing vasodilation and endothelial function. It engages proteins such as apolipoprotein B-100 in the inflammatory process and connects to pathways involving NF-kB influencing various cellular responses.

Oxidized phospholipids relate mainly to cardiovascular diseases and atherosclerosis. The accumulation and interaction of ox-LDL with cells in the arterial wall drive plaque formation and instability which can lead to myocardial infarction. Additionally these modified lipids link with diabetes-related complications due to their role in promoting vascular inflammation. Proteins like lectin-like oxidized LDL receptor-1 (LOX-1) mediate ox-LDL uptake emphasizing its damaging role in metabolic syndromes.
