Phospholipids containing choline are hydrolyzed by the PC Hydrolysis Enzyme. The choline is used by the Enzyme Mix IV along with the OxiRed Probe resulting in a colorimetric product with an absorbance at 570 nm and fluorometric excitation/emission at 535/587 nm.

Phospholipid Assay Kit (Colorimetric/Fluorometric) (ab234050) is a simple, accurate and reproducible way to measure total choline-containing phospholipids in biological samples such as serum and plasma.
This kit provides a high-throughput assay of inhibitors/inducers affecting phospholipid metabolism. The signal is directly proportional to the phospholipid concentration of phospholipids in the sample and can detect phospholipids in the range of 0.1 to 10 nmol per sample.
This product is manufactured by BioVision, an Abcam company and was previously called K351 Phospholipid Assay Kit (Colorimetric/Fluorometric). K351-100 is the same size as the 100 test size of ab234050.
Phospholipids are a major component of lipid bilayer membranes. In addition, phospholipids are biologically important molecules as they are closely involved in signal transduction. Most phospholipids contain one diglyceride, a phosphate group, and one choline moiety. In human serum and plasma, almost 95% of the phospholipids are choline-containing phospholipids such as phosphatidylcholine and sphingomyelin. The remaining 5% is mainly choline-free phospholipids (i.e. phosphatidylethanolamine and phosphatidylserine). Phospholipid concentration is considered a disease biomarker as it can be altered in many lung, liver and heart diseases.

Biological function summary
Phospholipids are vital in maintaining the integrity and functionality of membranes. They form lipid bilayers providing impermeable barriers for cells and organelles. Phospholipid molecules participate in cell signaling serving as precursors for signaling lipids like phosphatidylinositol. These molecules can organize into complex structures known as liposomes or micelles depending on their environment. In addition they interact extensively within membrane proteins affecting the protein's structure and function.
Pathways
Phospholipids play a significant role in the signal transduction and apoptosis pathways. In the signal transduction pathway phosphatidylinositol derivatives act as secondary messengers regulating various cellular responses. Proteins such as phospholipase C and kinase enzymes interact closely with these derivatives illustrating their involvement in complex cellular processes. The apoptosis pathway also relies on phospholipids with the release of ceramide from sphingomyelin degradation triggering programmed cell death. These interactions highlight important aspects of cellular regulation and homeostasis.
