Estimation of proline in local honey, human serum, spinach leaves lysate and rat liver lysate. Proline concentrations were 425.3 +/-48.9 mg/kg in local honey, 248.2+/-3.9 µM in human serum, 0.877 +/- 0.015 µmol/g-wet wt in spinach leaves and 13.3+/-1.4 pmol/µg protein in rat liver lysate.

Biological function summary
The cyclic structure of the proline amino acid influences protein folding and stability. Proline plays an important role in the formation of turns and loops within protein structures which are critical for creating functional three-dimensional configurations. Additionally proline residues often emerge as components of enzyme active sites influencing substrate specificity and enzyme activity. Although it is not part of any particular large molecular complex its presence within numerous proteins highlights its essential role in protein architecture and function.
Pathways
Proline integrates into both the proline metabolism pathway and collagen biosynthesis. In proline metabolism it contributes to the proline cycle affecting the cellular redox state by interconverting with pyrroline-5-carboxylate. Proline is involved in the synthesis of collagen a major structural protein in mammals. Its presence is linked with enzymes such as prolyl hydroxylase and proline dehydrogenase which play pivotal roles in these pathways. These connections highlight proline’s functional significance in maintaining cellular and tissue health.
