Human Striatal Medium Spiny GABAergic Neurons (MSNs) are derived from normal control iPSC line WC-30. BrainXell MSNs have high neuronal purity (>80%) and comprise predominantly inhibitory neurons. Labelling with the pan-neuronal marker MAP2 and the GABA-specific marker highlights the neuronal purity of these cells. Labelling for the medium spiny neuron marker, DARRP32 along with MAP2 confirms the presence of medium spiny neurons. MSNs are adherent and exhibit substantial neurite outgrowth within the first week in culture, showing the characteristic cell shape and extension of processes classic cortical GABAergic neurons. These neurons also demonstrate pronounced electrophysiological activity after two weeks in culture, as demonstrated by multi-electrode array (MEA) recordings.
Vials of MSNs are delivered with three supplements which are designed to facilitate the maturation and health of the cultures. BrainFast GABA is a seeding supplement which promotes cell survival at thaw and completes the differentiation process from late-stage neural progenitor to neuron. BrainFast SK enhances neuronal purity, and BrainFast D4 accelerates maturation.
