The ADRA2B also known as alpha-2B adrenergic receptor is a G protein-coupled receptor that interacts with catecholamines like norepinephrine and epinephrine. This receptor with a mass of around 54 kDa forms a part of the adrenergic receptor family which includes other members like the alpha-1 adrenergic receptor (ADRA1) and alpha-2A adrenergic receptor. The ADRA2B is mainly expressed in various tissues including adipose tissue the central nervous system and vascular smooth muscles playing a significant role in mediating several physiological responses.
The ADRA2B receptor facilitates vasoconstriction inhibiting neurotransmitter release in the sympathetic nervous system. It does not typically form part of a large protein complex but interacts with G proteins to mediate its cellular effects. This receptor also works by modulating levels of cyclic AMP within cells thereby influencing cellular signaling and response to stress stimuli.
The ADRA2B receptor largely contributes to the adrenergic signaling pathway which is important for the fight-or-flight response. Additionally it engages with related pathways such as the MAPK signaling pathway impacting cellular responses like proliferation and differentiation. Within these pathways it interacts with proteins like beta-adrenergic receptors coordinating with them to fine-tune cardiovascular and metabolic functions.
The ADRA2B receptor has associations with conditions like hypertension and heart failure. Its role in promoting vasoconstriction links it to elevated blood pressure where it frequently interacts with proteins like angiotensin II receptors. Additionally variations in the ADRA2B gene have appeared in research concerning post-traumatic stress disorder (PTSD) potentially due to its interaction with noradrenergic systems affecting stress hormone release.
