Bcl-2 Bcl-XL and Bcl2L2 are members of the Bcl-2 protein family known for their roles in regulating apoptosis through mitochondrial membrane integrity. Bcl-2 also known as B-cell lymphoma 2 has a mass of around 26 kDa and is expressed in several tissues including the lymphatic system brain and reproductive organs. Bcl-XL is similarly pivotal in the cellular life cycle whereas Bcl2L2 also called Bcl-w shares overlapping functions. These proteins primarily localize to the outer mitochondrial membrane where they control the release of apoptogenic factors.
Biological function summary

The Bcl-2 family proteins regulate programmed cell death by either promoting or inhibiting apoptosis. As anti-apoptotic members of this family Bcl-2 Bcl-XL and Bcl2L2 bind to pro-apoptotic proteins preventing them from forming pores in the mitochondrial membrane. They are part of a complex dynamic with other Bcl-2 family proteins balancing cell survival and death. These interactions impact cell proliferation differentiation and survival under stress conditions influencing normal development and immune responses.

Pathways

The Bcl-2 family proteins act within the intrinsic apoptotic pathway which centers on mitochondrial regulation. They modulate the pathway by interacting with pro-apoptotic proteins like Bax and Bak which facilitate mitochondrial outer membrane permeabilization. This pathway plays a role in cellular responses to DNA damage and cytokine signaling connecting to broader cellular mechanisms like the stress response pathway. Their anti-apoptotic action allows cells to survive despite internal cues for programmed death contributing to tissue homeostasis.

Defects in Bcl-2 Bcl-XL and Bcl2L2 proteins relate closely to cancer and autoimmunity.
