Biological function summary
The Bcl-2 family proteins regulate programmed cell death by either promoting or inhibiting apoptosis. As anti-apoptotic members of this family Bcl-2 Bcl-XL and Bcl2L2 bind to pro-apoptotic proteins preventing them from forming pores in the mitochondrial membrane. They are part of a complex dynamic with other Bcl-2 family proteins balancing cell survival and death. These interactions impact cell proliferation differentiation and survival under stress conditions influencing normal development and immune responses.
Pathways
The Bcl-2 family proteins act within the intrinsic apoptotic pathway which centers on mitochondrial regulation. They modulate the pathway by interacting with pro-apoptotic proteins like Bax and Bak which facilitate mitochondrial outer membrane permeabilization. This pathway plays a role in cellular responses to DNA damage and cytokine signaling connecting to broader cellular mechanisms like the stress response pathway.
