MEK1 MEK2 and MEK5 are important components in the MAPK/ERK signaling pathway while the Heme-regulated inhibitor (HRI) functions differently regulating protein synthesis. MEK proteins also referred to as MAP2Ks or MAPK/ERK kinases are dual-specificity kinases with MEK1 and MEK2 each having a molecular mass of about 44 kDa. They are broadly expressed in various tissues including brain and heart while MEK5 weighs around 47-49 kDa and shows expression in skeletal muscle. HRI also known as EIF2AK1 has an essential role in ensuring that cells do not produce excessive proteins and is mainly expressed in erythroid cells.
Biological function summary

MEK proteins are key components in cell signal transduction influencing numerous cellular processes such as proliferation differentiation and survival. MEK1 and MEK2 are closely related and often function together in signaling cascades while MEK5 activates ERK5 with distinct effects. Meanwhile HRI exerts control over protein translation by phosphorylating the eukaryotic initiation factor 2 alpha subunit (eIF2α) in response to stress conditions like iron deficiency. This inhibition prevents overproduction of globin proteins when heme is insufficient therefore balancing hemoglobin synthesis.

Pathways

MEK1 and MEK2 are integral parts of the MAPK/ERK pathway which plays an important role in growth factor signaling. They transmit signals from activated Ras protein eventually leading to ERK1/2 activation which influences gene expression. MEK1/2 inhibitors like AZD8330 can reduce downstream signaling which is useful in studying pathway dynamics. MEK5 participates in the MEK5-ERK5 pathway which sometimes shows cross-talk with MAPK/ERK signaling. HRI however is involved in pathways related to stress responses and does not overlap with these MAPK pathways.

MEK1 and MEK2 are implicated in various cancers particularly those with mutations leading to excessive MAPK pathway activation such as melanoma. Certain cancers exploit these mutations to drive tumor growth linking them tightly with cancer proteins like BRAF. In contrast HRI is connected to disorders of the red blood cells such as β-thalassemia. In this disorder effective HRI activity helps mitigate imbalances in globin synthesis interacting with other proteins involved in stress responses and erythropoiesis.
