Interferon beta Caspase-3 and Caspase-8 each play distinct mechanical roles in cellular processes. Interferon beta a type I interferon acts in antiviral defense and immune regulation with a mass of approximately 20 kDa. It is expressed in various cells such as fibroblasts and epithelial cells. Caspase-3 and Caspase-8 with masses of around 32 kDa and 55 kDa respectively are central executors in the apoptotic process. Caspase-3 also known as CPP32 and Caspase-8 frequently found in signals initiated by death receptors reside in the cytosol across different tissues.
Biological function summary

These proteins contribute to essential processes within the cell and immune system. Interferon beta plays a significant role in innate and adaptive immune responses by stimulating the antiviral state and regulating the activity of immune cells. It is not part of a complex. Caspase-3 acts as an executioner caspase cleaving cellular components to trigger apoptosis. Caspase-8 initiates the extrinsic apoptosis pathway by activating downstream caspases like Caspase-3 often forming part of the death-inducing signaling complex (DISC) upon activation by death receptors.

Pathways

Their biological roles fit into important signal cascades. Interferon beta operates within the JAK-STAT pathway modulating gene expression in response to pathogen invasion. Caspase-3 and Caspase-8 integrate into the apoptosis pathway. Caspase-8 connects the extrinsic pathway triggered by receptors like Fas (CD95) while Caspase-3 forms a node for executing apoptosis once activated either extrinsically or by the intrinsic pathway involving proteins like cytochrome c.

Interferon beta connects closely to autoimmune diseases like multiple sclerosis modulating immune attacks on the nervous system. Caspase-3 and Caspase-8 play roles in neurodegenerative diseases and cancer. In cancers defects in apoptosis often involve dysregulation of Caspase-8 affecting interactions with proteins such as FLIP. Caspase-3 through its role in apoptosis relates to programmed cell death pathway disruptions underlying neurodegeneration in conditions like Alzheimer's disease.
