Blooms Syndrome Protein also known as BLM is a RecQ helicase family member with a mass of approximately 141 kDa. This protein Tau and Estrogen Receptor among others also participate in repairing DNA and maintaining genomic stability. BLM is expressed in cells with high proliferative activity such as those in epithelial tissues. BLM plays a critical role in resolving DNA structures during replication and repair minimizing errors and maintaining fidelity. It acts alongside DNA polymerase iota and DNA polymerase eta to ensure proper DNA synthesis and repair.
Biological function summary

BLM and these associated proteins safeguard the integrity of the genome by participating in homologous recombination repair. BLM often functions in complex with topoisomerase IIIα and RMI1/RMI2 forming a complex to process recombination intermediates. TDP1 aids in this defensive mechanism by processing topoisomerase-induced DNA damage. The Estrogen Related Receptor alpha modulates gene expression influencing cellular responses to DNA damage while H-ERG contributes to the electrophysiological properties of cardiac tissues.

Pathways

BLM actively participates in the homologous recombination repair and the Fanconi Anemia pathway helping resolve replication fork blocks and breaks. In these pathways BLM closely interacts with proteins like Rad51 which assists in strand exchange and BRCA1/2 which participates in DNA repair.
