Supplementary information

RSK1 also known as p90 ribosomal S6 kinase or RPS6KA1 is a serine/threonine kinase with a mass of approximately 90 kDa. p90 RSK is expressed in various tissues showing a wide distribution pattern in mammals. S6K1 (p70 ribosomal S6 kinase) similarly acts as a serine/threonine kinase and is found predominantly in tissues such as the brain heart and skeletal muscle. ROCK1 and ROCK2 (Rho-associated coiled-coil containing kinases) are two closely related serine/threonine kinases with masses of about 158 kDa each found in almost all tissue types including smooth muscle and the brain.
Biological function summary

The RSK1 kinase regulates protein synthesis and cell proliferation by phosphorylating various substrates including the ribosomal S6 protein. This kinase connects with the MAPK/ERK signaling pathway and functions as a mediator of cellular response to extracellular signals. S6K1 a downstream effector of mTORC1 is important for protein translation and cell growth. ROCK1 and ROCK2 play key roles in actin cytoskeletal reorganization and are part of the Rho/ROCK complex influencing cell motility shape and apoptosis.

Pathways

RSK1 integrates into the MAPK/ERK pathway where it contributes to the phosphorylation of substrates like CREB and other transcription factors driving gene expression changes. S6K1 aligns with the mTOR pathway where it is vital for the regulation of cellular metabolism and growth. Within the Rho/ROCK pathway ROCK1 and ROCK2 influence the cytoskeleton interfacing with proteins such as LIMK1 and cofilin modulating actin dynamics.

RSK1 has been linked to breast cancer as its expression modulates the proliferation of cancer cells. This has implications for therapeutic interventions targeting the MAPK/ERK pathway. S6K1's connection to type 2 diabetes stems from its role in insulin resistance via the mTOR pathway. The association of ROCK1 and ROCK2 with cardiovascular diseases arises from their function in smooth muscle contraction and vascular homeostasis potentially connecting with other proteins such as E-cadherin which alters endothelial cell functions.
