Integrins are transmembrane proteins composed of alpha and beta subunits that mediate cell adhesion and signaling. The specific integrins including Integrin alpha 2 Integrin beta 1 Integrin alpha 3 Integrin alpha 5 CD41 and Integrin beta 3 are well known for their roles in binding to extracellular matrix proteins facilitating cell-to-matrix interactions. These integrins are expressed in various human tissues and cell types. The mass of these integrins varies; for example Integrin alpha 2 is approximately 150 kDa. These proteins go by alternate names such as VLA-2 (very late antigen 2) and GPIIb/IIIa for CD41 and Integrin beta 3 respectively.
Biological function summary

Integrins participate in key cellular functions like migration proliferation and survival. They are parts of larger protein complexes at the cell surface acting together with other proteins to transmit signals inside the cell. Integrins specifically engage with extracellular matrix components like fibronectin and collagen influencing cellular responses to the external environment. The GRGDS peptide known to mimic fibronectin binding sites serves as an inhibitor to some integrin-mediated interactions by competing with natural ligands.

Pathways

Integrins are critical components in integrin-mediated signaling pathways and the cellular response to fibronectin. These pathways involve interactions with signaling proteins such as focal adhesion kinase (FAK) and Src family kinases which facilitate signal transduction from the extracellular matrix into the cell interior. Such pathways modulate cytoskeleton organization and gene expression. The integrin fibronectin binding is particularly significant in pathways that regulate cell shape and motility and is connected to Rac and Rho GTPases by downstream signaling cascades.

Abnormal integrin function relates to cancer metastasis and thrombosis. In cancer integrin signaling can promote tumor cell invasion and metastasis often through alterations in cell adhesion and migration dynamics. CD41 part of the glycoprotein IIb/IIIa complex when defective is implicated in bleeding disorders such as Glanzmann's Thrombasthenia. These dysfunctions demonstrate a direct link between integrin expression patterns subtype interactions and disease progression relating integrins closely with a host of pathological conditions.
