SPHK1 also known as sphingosine kinase 1 VCP or valosin-containing protein and 5-lipoxygenase (5-LO) are important enzymes in cellular signaling. SPHK1 catalyzes phosphorylation of sphingosine into sphingosine-1-phosphate (S1P) a lipid signaling molecule. It is expressed in various tissues including the liver kidney and lung. SPHK1 has a molecular weight of around 43 kDa. VCP a 90 kDa protein is a member of the ATPase family associated with various cellular activities and it is found throughout many cell types. 5-LO with a molecular weight of approximately 78 kDa is responsible for leukotriene synthesis from arachidonic acid and mainly expresses in immune cells like leukocytes.
Biological function summary

SPHK1 plays an important role in the regulation of cell growth survival and migration. It takes part in multiple cellular functions through the production of S1P. VCP involved in various cellular processes acts in protein degradation membrane fusion and DNA damage response. It interacts with other proteins to form complexes necessary for these functions. 5-LO contributes to inflammatory responses and regulates immune reactions by producing leukotrienes important mediators of inflammation.

SPHK1 and its product S1P are vital components of the sphingolipid signaling pathway influencing cell proliferation and apoptosis. This pathway interacts with other pathway molecules like Akt and ERK linking SPHK1 with important signaling networks. VCP is an integral part of the ubiquitin-proteasome pathway maintaining protein homeostasis in the cell through interactions with ubiquitin ligases. 5-LO integrates into the arachidonic acid metabolism pathway and connects with proteins such as FLAP which assist in leukotriene production.

SPHK1 has strong associations with cancer progression and metastasis due to its role in cell survival signaling. In cancer cells SPHK1 activity relates to increased growth and resistance to programmed cell death often interacting with oncogenic pathways like PI3K/Akt.
