SFRP1 also known as secreted frizzled-related protein 1 functions as a modulator by binding to Wnt ligands and preventing them from interacting with Frizzled receptors. This protein with a molecular weight of approximately 35 kDa exhibits broad expression in various tissues such as the heart kidney and uterus. Its role is significant in the extracellular matrix where it can regulate cellular processes by affecting the Wnt signaling pathway. Wnt3 part of the Wnt family shares similar involvement in signaling pathways but specifically contributes to early developmental stages.
Biological function summary

SFRP1 and Wnt3 modulate processes critical for cellular growth and differentiation by interacting with Wnt signaling components. SFRP1 inhibits the Wnt pathway by sequestering Wnt proteins away from receptors therefore balancing the signal transduction. Wnt3 activates signaling through its binding to Frizzled proteins facilitating downstream effects essential for embryonic development. Pathways

Conversely Wnt3 positively influences this pathway promoting β-catenin accumulation and gene expression important for tissue development. These proteins also connect with the planar cell polarity (PCP) pathway where they coordinate cellular orientation processes. β-Catenin and Frizzled receptors serve as partners within these pathways showcasing the complex interplay in cellular signaling.

Alterations in SFRP1 and Wnt3 activity link to various conditions. SFRP1 often exhibits decreased expression in several cancers including breast and lung cancer suggesting its role in tumor suppression. Disregulation in SFRP1 can interfere with cell proliferation and apoptosis highlighting connections to oncogenic pathways. Wnt3 associations with bone disorders like osteoporosis arise due to its role in bone density regulation. In these cases interacting partners like Dkk1 (Dickkopf-related protein 1) and LRP5 involved in bone homeostasis also display altered expressions further complicating disease outcomes.
