Upon arrival, the vial should be stored in liquid nitrogen vapor phase and not at -80°C. Storage at -80°C may result in loss of viability.
1. Thaw the vial in 37°C water bath for approximately 1-2 minutes.
2. Transfer the cell suspension (0.8 mL) to a 15 mL/50 mL conical sterile polypropylene centrifuge tube containing 8.4 mL pre-warmed culture medium, wash vial with an additional 0.8 mL culture medium (total volume 10 mL) to collect remaining cells, and centrifuge at 201 x g (rcf) for 5 minutes at room temperature. 10 mL represents minimum recommended dilution. 20 mL represents maximum recommended dilution.
3. Resuspend the cell pellet in 5 mL pre-warmed culture medium and count using a haemocytometer or alternative cell counting method seed all remaining cells into a T25.
4. Incubate the culture at 37°C incubator with 5% CO2. Check the culture one day after revival and continue to check until 80% confluent. Media change can be given if needed.
5. Once confluent passage into an appropriate flask at a density of 2x104 cells/cm2. Seeding density is given as a guide only and should be scaled to align with individual lab schedules. Cultures should be monitored daily.
We will provide viable cells that proliferate on revival.

The 5HT3D Receptor also known as 5-Hydroxytryptamine Receptor 3D is a subtype of serotonin receptor. It is part of the ligand-gated ion channel family. This receptor is involved in mediating fast synaptic transmission in the central and peripheral nervous systems. The 5HT3D Receptor has a molecular mass of approximately 56 kDa. Expression of this receptor occurs in human brains and the gastrointestinal tract where it performs its role in neuronal signaling.
The 5HT3D Receptor functions as a component of the ligand-gated ion channel complex allowing the passage of cations including sodium (Na+) and calcium (Ca2+) upon serotonin binding. This action affects neuronal excitability and neurotransmitter release. The receptor plays a significant role in intestinal motility modulating gut motility and in the central nervous system where it contributes to emotions and cognitive processes.
The 5HT3D Receptor is involved in the serotonergic synapse pathway which is important for transmitting signals in the nervous system. The receptor works alongside other serotonin receptors such as 5HT3A influencing neurotransmitter release and neuronal excitability. It also interacts within the GABAergic synapse pathway where it may affect inhibitory neurotransmission alongside the GABA receptors. These interactions can impact mood regulation and cognitive functions.
The 5HT3D Receptor has associations with conditions such as irritable bowel syndrome (IBS) and anxiety disorders. Abnormal expression or function of this receptor can disrupt gastrointestinal motility leading to IBS symptoms. In the context of anxiety disorders the receptor’s interaction with neurotransmitter pathways can influence anxiety levels. Additionally it connects with other proteins such as serotonin transporter (SERT) and GABA receptors which are implicated in anxiety regulation.
