Handling procedures

Initial handling guidelines

Upon arrival, the vial should be stored in liquid nitrogen vapor phase and not at -80°C. Storage at -80°C may result in loss of viability.

1. Thaw the vial in 37°C water bath for approximately 1-2 minutes.
2. Transfer the cell suspension (0.8 mL) to a 15 mL/50 mL conical sterile polypropylene centrifuge tube containing 8.4 mL pre-warmed culture medium, wash vial with an additional 0.8 mL culture medium (total volume 10 mL) to collect remaining cells, and centrifuge at 201 x g (rcf) for 5 minutes at room temperature. 10 mL represents minimum recommended dilution. 20 mL represents maximum recommended dilution.
3. Resuspend the cell pellet in 5 mL pre-warmed culture medium and count using a haemocytometer or alternative cell counting method seed all remaining cells into a T25.
4. Incubate the culture at 37°C incubator with 5% CO2. Check the culture one day after revival and continue to check until 80% confluent. Media change can be given if needed.
5. Once confluent passage into an appropriate flask at a density of 2x104 cells/cm2. Seeding density is given as a guide only and should be scaled to align with individual lab schedules. Cultures should be monitored daily.

Subculture guidelines
  • All seeding densities should be based on cell counts gained by established methods.
  • A guide seeding density of 2x104 cells/cm2 is recommended.
  • Cells should be passaged when they have achieved 80-90% confluence.
  • Do not allow the cell density to exceed 7x104 cells/cm2.

NLRC5 also known as NOD-like receptor family CARD domain-containing 5 is an important protein in the immune response. It has a molecular mass of approximately 186 kDa. This protein locates primarily in the cytoplasm of cells with high expression levels in immune tissues such as the spleen and lymph nodes. Mechanically NLRC5 acts as a transcriptional regulator controlling the expression of major histocompatibility complex (MHC) class I genes. This function plays a critical role in the immune system by facilitating the presentation of endogenous antigens.
Biological function summary

NLRC5 engages in enhancing immune surveillance and antiviral responses by regulating MHC class I transcription. It acts as part of the larger NLR family of proteins which are known to participate in the immune system's response to pathogens. In addition to its role in the immune response NLRC5 can interact with components of the inflammasome complex although its presence in the inflammasome remains controversial. These mechanisms are essential in initializing and sustaining an immune response against infections.

Pathways

NLRC5 involves itself in the interferon signaling pathway and antigen processing and presentation pathways. In the context of interferon signaling NLRC5 influences the activation of genes that drive antiviral states by interacting with proteins such as IRF1 and STAT1. This regulatory role positions NLRC5 as a bridge between innate and adaptive immune responses ensuring proper signaling and presentation of antigens to T cells for effective immune resolution.

NLRC5 exhibits alterations in conditions such as some auto-inflammatory disorders and cancer. Deregulation of NLRC5 can contribute to diseases like systemic lupus erythematosus where its altered expression affects immune function and tolerance. In certain cancers such as hepatocellular carcinoma NLRC5 interacts with oncogenic pathways influencing tumor growth and immune evasion. Researchers explore connections between NLRC5 and proteins like MHC class I molecules to understand their roles in disease mechanisms and therapeutic potentials.
