Handling procedures

Initial handling guidelines

Upon arrival, the vial should be stored in liquid nitrogen vapor phase and not at -80°C. Storage at -80°C may result in loss of viability.

1. Thaw the vial in 37°C water bath for approximately 1-2 minutes.
2. Transfer the cell suspension (0.8 mL) to a 15 mL/50 mL conical sterile polypropylene centrifuge tube containing 8.4 mL pre-warmed culture medium, wash vial with an additional 0.8 mL culture medium (total volume 10 mL) to collect remaining cells, and centrifuge at 201 x g (rcf) for 5 minutes at room temperature. 10 mL represents minimum recommended dilution. 20 mL represents maximum recommended dilution.
3. Resuspend the cell pellet in 5 mL pre-warmed culture medium and count using a haemocytometer or alternative cell counting method seed all remaining cells into a T25.
4. Incubate the culture at 37°C incubator with 5% CO2. Check the culture one day after revival and continue to check until 80% confluent. Media change can be given if needed.
5. Once confluent passage into an appropriate flask at a density of 2x104 cells/cm2. Seeding density is given as a guide only and should be scaled to align with individual lab schedules. Cultures should be monitored daily.

Subculture guidelines
  • All seeding densities should be based on cell counts gained by established methods.
  • A guide seeding density of 2x104 cells/cm2 is recommended.
  • Cells should be passaged when they have achieved 80-90% confluence.

Rab2 also known as Ras-related protein Rab-2 is a small GTPase with a mass of approximately 24 kDa. It belongs to the Rab family of proteins which are key regulators of intracellular membrane trafficking. Rab2 plays a critical role in vesicle transport between the endoplasmic reticulum (ER) and the Golgi apparatus. It is widely expressed in various tissues with significant presence in brain and liver tissues. This expression pattern indicates its involvement in essential cellular processes across different cell types.
Biological function summary

Ras-related protein Rab-2 mediates the trafficking of proteins and lipids within the secretory pathway. Rab2 associates with the cytosolic face of membranes aiding in the formation and movement of vesicles. It forms part of a complex that regulates tethering and docking facilitating vesicle fusion. Rab2 interaction with its effectors ensures accurate delivery of cargo to the correct cellular compartments. This functionality supports the maintenance of cellular organization and communication.

Pathways

Ras-related protein Rab-2 functions in the ER-Golgi transport pathway and the autophagy pathway. In the ER-Golgi transport pathway it interacts with proteins such as Golgi SNAREs to enhance vesicle sorting and fusion. Rab2 also has connections with the autophagy-related proteins where it supports the formation of autophagosomes therefore contributing to cellular homeostasis and turnover. Its activity impacts the coordination between membrane trafficking and recycling processes within cells.

Ras-related protein Rab-2 has relevance to neurodegenerative conditions and cancer. Abnormal Rab2 expression or function associates with Alzheimer's disease where its dysregulation affects ER-Golgi transport and protein processing. Additionally Rab2 involvement in cancer relates to its role in protein trafficking and cellular growth interacting with proteins like Ras and RalA within signaling cascades. These interactions highlight the importance of Rab2 in the pathology and progression of these diseases.
