Annexin A3 also known as ANXA3 is a 36 kDa protein that belongs to the annexin family. This family is known for calcium-dependent phospholipid-binding activity. ANXA3 specifically binds to phosphatidylserine a type of phospholipid found in cell membranes. It is expressed in various tissues with notable presence in myeloid cells particularly in the bone marrow and granulocytes.
Biological function summary

ANXA3 plays roles in membrane-related events including vesicle trafficking and exocytosis. It is not part of a large complex but associates with other proteins depending on its cellular function. ANXA3 can also regulate inflammation by modulating macrophage activation and may influence signal transduction pathways that are linked to immune responses.

Pathways

ANXA3 functions in both inflammation and apoptosis. It is closely related to the apoptosis pathway by interacting with signaling molecules like caspases which are critical for the execution of cell death. Additionally through its action in the inflammation pathway ANXA3 is known to interact with proteins such as NF-κB an important regulator of immune and inflammatory responses.

Alterations in ANXA3 expression or function associate with various conditions most notably cancer and autoimmune diseases. For instance in certain cancers ANXA3 may contribute to tumor growth and metastasis. In autoimmune diseases its regulation of macrophage activity can misbalance immune responses leading to pathogenesis. Proteins such as ANXA1 and S100A11 also play roles in these pathways and may function along with ANXA3 in disease progression.
