Lane 1 : Wild-type HeLa cell lysate (20 ug)

ab181092 was shown to specifically react with Actin Regulatory Protein CAPG/MCP in wild-type HeLa cells. Loss of signal was observed when knockout cell line ab265766 (knockout cell lysate ab257378) was used. Wild-type and Actin Regulatory Protein CAPG/MCP knockout samples were subjected to SDS-PAGE. ab181092 and Anti-GAPDH antibody [6C5] - Loading Control (ab8245) were incubated at room temperature for 2.5 hours at 1 in 1000 dilution and 1 in 20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (ab216776) secondary antibodies at 1 in 20000 dilution for 1 hour at room temperature before imaging.

Biological function summary
This actin regulatory protein serves as an important player in the remodeling of the actin cytoskeleton. CAPG functions by capping the fast-growing ends of actin filaments which influences cell motility and structure. It is not known to be part of a larger protein complex but interacts closely with other actin-modulating proteins. MCP's regulation of cytoskeletal dynamics allows cells to respond swiftly to environmental changes assisting cellular processes like migration and secretion.
Pathways
CAPG is intricately involved in pathways controlling cell motility and structure including the cytoskeletal rearrangement pathway. It works alongside proteins such as cofilin and Arp2/3 complex which also regulate actin dynamics. These proteins together facilitate processes like cell division and migration making their interactions essential for proper cellular function.
