Frizzled homolog 1 (FZD1) also known as Fz-1 is a member of the Frizzled family of G protein-coupled receptor proteins involved in the Wnt signaling pathway. FZD1 has a molecular mass of approximately 74 kDa. It consists of seven transmembrane domains and is found mainly on the cell surface. Expression of FZD1 occurs in various tissues including the kidney heart liver and lung. The target acts as a receptor for Wnt proteins facilitating the binding and signaling required for downstream effects.
Biological function summary

FZD1 participates in important processes concerning cell development differentiation and growth. As a member of the Frizzled family FZD1 often forms a functional complex with LRP5/6 co-receptors upon Wnt ligand binding. This complex initiates intracellular signaling cascades that lead to the regulation of target genes. In addition to its role in development FZD1 also has a major impact on maintaining stem cell renewal and tissue homeostasis.

Pathways

FZD1 functions significantly in both the canonical and non-canonical Wnt signaling pathways. In the canonical pathway FZD1 interacts with proteins such as Disheveled (DVL) and β-catenin leading to alterations in gene expression that influence cellular proliferation and differentiation. In the non-canonical pathway FZD1 also associates with other proteins including Rho and JNK which contribute to changes in cell polarity and motility. These pathways highlight FZD1's involvement in complex networks essential for cellular and tissue dynamics.

FZD1 has associations with conditions like cancer and osteoporosis. Aberrations in FZD1 expression or function can lead to disrupted Wnt signaling contributing to tumorigenesis by affecting cell proliferation and survival pathways. FZD1's role in bone development ties it to osteoporosis where improper signaling affects bone density and strength. Moreover FZD1 associates with other proteins such as DVL and β-catenin in these pathological processes highlighting its impact on disease progression.
