Glypican 4 also known as GPC4 is a heparan sulfate proteoglycan a member of the glypican family. This protein is anchored to the cell membrane and plays a role in cellular processes by attaching to the cell surface via a glycosylphosphatidylinositol (GPI) anchor. Its mass is approximately 60-70 kDa. Glypican 4 is expressed in various tissues including the brain pancreas kidney and skeletal muscle. In these tissues Glypican 4 is involved in cell communication and modulation of signaling activities at the cell surface.
Biological function summary

Glypican 4 influences cell growth differentiation and morphogenesis. It interacts with extracellular ligands and growth factors playing a role in developmental processes. Glypican 4 is often part of a complex with other signaling molecules including Wnt and fibroblast growth factors. These interactions help regulate processes essential for organ development and tissue patterning. Glypican 4 through these molecular associations influences signaling pathways that affect both cellular architecture and function.

Pathways

Glypican 4 plays a role in Wnt signaling and Hedgehog pathways. These pathways are critical for embryonic development and cell fate determination. Within the Wnt signaling pathway Glypican 4 interacts with the Wnt protein modulating its distribution and activity on the cell surface. In the Hedgehog pathway Glypican 4 helps modulate hedgehog proteins like Sonic Hedgehog facilitating their movement and interaction with cell surface receptors.

Glypican 4 is connected to some cancers and metabolic disorders like diabetes. Overexpression or mutations in Glypican 4 can contribute to oncogenesis as it may lead to aberrant activation of signaling pathways making it relevant in cancers like pancreatic and brain tumors. Additionally Glypican 4's role in metabolic pathways links it to insulin-related pathways emphasizing its connection with metabolic disorders such as Type 2 diabetes.
