Lysate preparation: Our lysates are made using RIPA buffer to which we add a protease inhibitor cocktail and phosphatase inhibitor cocktail (ratio: 300:100:10). This means that the protein of interest is denatured. If you require a native form of the protein please use the live cell version. Please refer to our lysis protocol for further details on how our lysates are prepared.

User storage instructions: Lyophilizate may be stored at 4°C. After reconstitution, store at -20°C for short-term storage or -80°C for long-term storage.

Activity summary

PI 3 Kinase Class 2A also known as Cpk is a significant enzyme involved in the regulation of various cellular processes. The target weighing approximately 170 kDa acts as a lipid kinase that phosphorylates phosphatidylinositol converting PI to PI3P. Cpk expresses mainly in tissues like the liver muscle and heart where it contributes to cellular homeostasis and metabolic regulation.

Biological function summary

This enzyme family modulates intracellular signaling cascades influencing processes such as cell growth survival and metabolism. Cpk forms part of a larger complex that interacts with adaptor proteins facilitating its recruitment to cellular membranes. This interaction is important for its signaling activity and function in maintaining cellular equilibrium.

Pathways

PI 3 Kinase Class 2A plays an integral role in the insulin signaling pathway and the TOR signaling pathway. In the insulin pathway it collaborates with proteins like Akt to modulate glucose uptake and storage in cells. Similarly in the TOR pathway Cpk contributes to nutrient sensing and cell growth regulation interconnected with proteins such as mTOR affecting protein synthesis and autophagy.

Dysregulation of PI 3 Kinase Class 2A links to cancer and type 2 diabetes. Alterations in its activity may lead to abnormal cell proliferation and survival characteristics of cancerous transformations associated with proteins like PTEN. Meanwhile its role in glucose metabolism ties it to insulin resistance and diabetes pathogenesis where interactions with insulin receptor substrates become important for disease progression.
