PIP4K2 alpha also known as phosphatidylinositol-5-phosphate 4-kinase type-2 alpha is an enzyme that catalyzes the phosphorylation of phosphatidylinositol-5-phosphate (PI5P) to produce phosphatidylinositol-45-bisphosphate (PI(45)P2). This protein has a mass of approximately 46 kDa. It is expressed in various tissues with high levels in the brain heart and kidney. PIP4K2 alpha plays a role in the regulation of intracellular signaling pathways through its enzymatic activity.
Biological function summary

PIP4K2 alpha participates in the regulation of phosphoinositide signaling and lipid metabolism. It contributes to the modulation of various cellular processes including cell growth and survival. PIP4K2 alpha doesn't form a large complex but interacts with other kinases to influence signaling pathways. This protein acts as a regulator of PI(45)P2 levels balancing phosphatidylinositol lipid signaling in cells.

Pathways

PIP4K2 alpha plays important roles in the phosphatidylinositol signaling pathway and the insulin signaling pathway. In the phosphatidylinositol signaling pathway PIP4K2 alpha works alongside other kinases like PIP5K1A to regulate the synthesis of PI(45)P2. In the insulin signaling pathway PIP4K2 alpha contributes to the regulation of glucose uptake and metabolism maintaining cellular energy balance.

Disruptions in PIP4K2 alpha activity have been associated with metabolic disorders such as diabetes and cancer. Altered levels of PI(45)P2 due to dysregulation of PIP4K2 alpha can lead to insulin resistance which is a hallmark of type 2 diabetes. Additionally due to its role in cellular growth changes in PIP4K2 alpha function can contribute to cancer cell proliferation where it may interact with proteins like AKT that are pivotal in tumor growth pathways.
