SFRP1 KO cell lysate available now. KO validated by Western blot. Free of charge wild type control included. Homozygous CRISPR/Cas9 KO: the 52 bp deletion in exon 2 leads to a truncated SFRP1 (tSFRP1) rather than a complete loss of protein. tSFRP1 is not secreted out of the cell, in contrast with WT SFRP1.

REACH authorisation
Abcam has not and does not intend to apply for the REACH Authorisation of customers' uses of products that contain European Authorisation list (Annex XIV) substances.
It is the responsibility of our customers to check the necessity of application of REACH Authorisation, and any other relevant authorisations, for their intended uses.

Lysate preparation: Our lysates are made using RIPA buffer to which we add a protease inhibitor cocktail and phosphatase inhibitor cocktail (ratio: 300:100:10). This means that the protein of interest is denatured. If you require a native form of the protein please use the live cell version. Please refer to our lysis protocol for further details on how our lysates are prepared.

User storage instructions: Lyophilizate may be stored at 4°C. After reconstitution, store at -20°C for short-term storage or -80°C for long-term storage.

This product is subject to limited use licenses from The Broad Institute and ERS Genomics Limited, and is developed with patented technology. For full details of the limited use licenses and relevant patents please refer to our limited use license and patent pages.

Supplementary information

SFRP1 also known as Secreted Frizzled-Related Protein 1 is a protein that functions mechanically by modulating Wnt signaling pathways. This protein weighs approximately 35 kiloDaltons. SFRP1 acts as a modulator by binding to Wnt ligands preventing them from interacting with their cell surface receptors. It is expressed in various tissues including the kidney retina lung and heart.
Biological function summary

The secreted glycoprotein SFRP1 plays a role in cellular processes such as cell growth differentiation and apoptosis. As a member of the soluble frizzled-related proteins it acts to inhibit Wnt signaling pathways which are critical in maintaining cellular homeostasis. SFRP1 commonly interacts in the extracellular environment where it forms part of a regulatory complex that includes other extracellular matrix components to achieve its function.

Pathways

The role SFRP1 plays is important in the Wnt signaling pathway and the regulation of β-catenin activity. These pathways are involved in embryonic development and tissue regeneration. SFRP1 directly interacts with other proteins like frizzled receptors and Wnt ligands to maintain balance in signaling. This modulation further influences gene expression patterns and cellular behavior.

An imbalance or alteration in SFRP1 activity is associated with cancer and osteoporosis. Its underexpression or lack of function can lead to abnormal cellular proliferation which is linked to tumorogenesis. In cancer SFRP1's interaction with the Wnt signaling pathway and its related proteins such as β-catenin highlights its role in maintaining normal cell function. Abnormal regulation also impacts bone density where SFRP1's regulatory function on Wnt S signaling may affect bone formation and lead to osteoporosis.
