REACH authorisation
Abcam has not and does not intend to apply for the REACH Authorisation of customers' uses of products that contain European Authorisation list (Annex XIV) substances.
It is the responsibility of our customers to check the necessity of application of REACH Authorisation, and any other relevant authorisations, for their intended uses.

Lysate preparation: Our lysates are made using RIPA buffer to which we add a protease inhibitor cocktail and phosphatase inhibitor cocktail (ratio: 300:100:10). This means that the protein of interest is denatured. If you require a native form of the protein please use the live cell version. Please refer to our lysis protocol for further details on how our lysates are prepared.

User storage instructions: Lyophilizate may be stored at 4°C. After reconstitution, store at -20°C for short-term storage or -80°C for long-term storage.

This product is subject to limited use licenses from The Broad Institute and ERS Genomics Limited, and is developed with patented technology. For full details of the limited use licenses and relevant patents please refer to our limited use license and patent pages.

Supplementary information

SH2D4A also known as Src homology 2 domain-containing protein 4A is a protein involved in signal transduction pathways. It has a molecular mass of approximately 58 kDa. This protein contains an SH2 domain which plays a role in mediating protein-protein interactions by binding to phosphotyrosine-containing motifs. SH2D4A is expressed in various tissues with higher levels observed in immune-related tissues including the spleen and lymph nodes. This expression pattern suggests a functional role in cellular communication and response mechanisms.
Biological function summary

The SH2D4A protein participates in the regulation of immune responses. It is not considered part of a large complex but interacts with other signaling molecules to influence downstream signaling cascades. By engaging with phosphorylated partners SH2D4A modulates pathways important for cell growth proliferation and immune activation. This function positions SH2D4A as an important mediator in cellular signaling networks underpinning immune system function.

Pathways

Research identifies SH2D4A as influencing both the MAPK and JAK-STAT pathways. It interacts with key proteins within these pathways such as MAPK1 which is integral for transmitting extracellular signals into cellular responses. These pathways regulate processes essential for cell division growth and development. SH2D4A's interaction with signaling components supports its role in controlling cellular responses to external stimuli and maintaining cellular homeostasis.

SH2D4A has been linked to conditions impacting the immune system. Abnormalities in this protein's function have correlations with autoimmune diseases and certain types of lymphomas. SH2D4A is connected to proteins like STAT5 which is important in cytokine signaling and immune cell differentiation. Dysregulation of SH2D4A leading to altered interaction with signaling molecules may drive pathological changes seen in these immune-related disorders.
