Lysate preparation: Our lysates are made using RIPA buffer to which we add a protease inhibitor cocktail and phosphatase inhibitor cocktail (ratio: 300:100:10). Activity summary

The Yes1 protein also known as p61-Yes is a member of the Src family kinases (SFKs) and has a molecular mass of approximately 61 kDa. This non-receptor tyrosine kinase engages in phosphorylation of tyrosine residues playing a significant role in cell signaling. It is widely expressed in many tissues including the brain heart and lungs suggesting its involvement in varied physiological processes.

Biological function summary

The Yes1 protein participates in cellular processes such as cell growth survival and differentiation. It contributes to signaling cascades that regulate these functions often working as part of protein complexes. Its ability to interact with various signaling molecules positions it as an important player in the control of cell behavior particularly in response to extracellular stimuli.

    Anti-Yes1 antibody [EPR3173] ab109265 Anti-Yes1 antibody [EPR3173] was shown to specifically react with Yes1 in wild-type HeLa cells in western blot. The band observed in the knockout cell line Human YES1 knockout HeLa cell line ab265202 (knockout cell lysate ab258290) lane below 60kDa may represent truncated forms and cleaved fragments. This has not been investigated further. Wild-type and Yes1 knockout samples were subjected to SDS-PAGE. Membrane was blocked for 1 hour at room temperature in 0.1% TBST with 3% non-fat dried milk. Anti-Yes1 antibody [EPR3173] ab109265 and Anti-GAPDH antibody [6C5] - Loading Control (Anti-GAPDH antibody [6C5] - Loading Control ab8245) were incubated overnight at 4°C at 1 in 1000 and 1 in 20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed ab216776) secondary antibodies at 1 in 20000 dilution for 1 hour at room temperature before imaging.
