Sandwich ELISA kits are designed for the specific detection and quantification of an antigen between two layers of antibodies, hence the name 'sandwich'. The first layer, a capture antibody, is coated on the assay plate. Samples containing the target antigen are added, and any antigen present binds to this antibody. A detection antibody follows, forming a complex with the antigen. This antibody is linked to an enzyme or a detectable tag, and upon adding a substrate, a detectable signal is produced. The intensity of this signal is directly proportional to the amount of antigen in the sample.
