Product details

Abcam's 11-dehydro-TXB2 in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of 11-dehydro-TXB2 in tissue culture media and urine.

A goat anti-rabbit IgG antibody has been precoated onto 96-well plates. Standards or test samples are added to the wells, along with an alkaline phosphatase (AP) conjugated-11-dehydro-TXB2 antigen and a polyclonal rabbit antibody specific to 11-dehydro-TXB2. After incubation the excess reagents are washed away. pNpp substrate is added and after a short incubation the enzyme reaction is stopped and the yellow color generated is read at 405 nm. The intensity of the yellow coloration is inversely proportional to the amount of 11-dehydro-TXB2 captured in the plate.

11-dehydro-Thromboxane B2 (11-dehydro-TXB2) is a major metabolite of Thromboxane B2 (TXB2) found in urine and plasma. It is produced by the dehydrogenation of the alcohol group on the C11 position of TXB2 by the enzyme 11-OH-dehydrogenase. The detection of 11-dehydro-TXB2 is often used to measure thromboxane production in vivo. Monitoring levels of this metabolite is a helpful tool in the study of several diseases, such as liver cirrhosis, cystic fibrosis, mastocytosis, systemic lupus erythematosus, thrombosis diseases, and other diseases involving platelet activation.11-dehydro-TXB2 has also been used in diabetes and asthma studies.

11-dehydro-TXB2 is a stable metabolite of thromboxane B2 (TXB2) formed by the action of thromboxane synthase. It is often used as a marker to measure thromboxane production in scientific studies. 11-dehydro-TXB2 has a molecular mass of approximately 368.5 g/mol. This compound is generally detected in urine where its levels reflect the overall thromboxane A2 production in the body. The conversion from TXB2 occurs rapidly making it a critical analyte in the study of thromboxane activity.
Biological function summary

11-dehydro-TXB2 helps assess platelet function and plays an integral role in understanding thrombotic processes. This metabolite is not part of a larger protein complex but it is an important player in systems where platelet aggregation and vasoconstriction occur. 11-dehydro-TXB2 is frequently used in research to investigate cardiovascular health providing insights into the regulation and disruption of normal platelet function.

Pathways

11-dehydro-TXB2 is a significant part of the arachidonic acid metabolism pathway. This pathway involves the conversion of arachidonic acid into various eicosanoids including thromboxane A2 which is quickly converted to TXB2 and subsequently to 11-dehydro-TXB2. Related proteins in this pathway include cyclooxygenase enzymes (COX-1 and COX-2) which facilitate the initial stages of eicosanoid synthesis.

11-dehydro-TXB2 is closely linked to cardiovascular diseases such as myocardial infarction and stroke. Elevated levels of 11-dehydro-TXB2 often indicate increased thromboxane production correlating with heightened thrombotic risk. In these disorders the metabolite serves as a biomarker for the effectiveness of antiplatelet therapy such as aspirin which targets and reduces thromboxane A2 synthesis through the inhibition of COX enzymes.
