4-Hydroxynonenal ELISA Kit is a competitive ELISA kit to measure 4-HNE in plasma, tissue Homogenate, serum, other biological fluids with a sensitivity of 18.75 pg/mL.

How the assay works

The concentration of the 4-HNE in the samples can determined by comparing the optical density to the standard curve. The OD is inversely proportional to the amount of 4-HNE captured from the samples.

Assay Specificity

No significant cross-reactivity or interference between 4-HNE and analogues was observed.

4-Hydroxynonenal ELISA Kit ab287803 protocol summary:

1. Wash plate 2 times before adding standard, sample and control wells
2. Add each standards or samples into appropriate wells. Immediately add Biotin-labeled Antibody Working Solution into each well.
3. Incubate for 45 minutes at 37°C.
4. Wash
5. Add SABC working solution into each well, cover the plate and incubate at 37ºC for 30 min
6. Wash
7. Add TMB substrate into each well, cover the plate and incubate at 37 ºC in dark within 15-30 min.
8. Add Stop solution and read results immediately at 450 nm.

4-Hydroxynonenal also known as 4-HNE is a reactive aldehyde formed during lipid peroxidation particularly from omega-6 polyunsaturated fatty acids like arachidonic acid. It has a relatively low molecular weight typically around 156.23 g/mol. Cells express 4-HNE under conditions of oxidative stress and it is widespread in tissues where high oxidative processes occur. This compound readily forms adducts with proteins lipids and DNA impacting cellular structure and signaling.
Biological function summary

4-Hydroxynonenal acts as a toxic lipid peroxidation product that modifies proteins and influences cell signaling pathways. It serves as a signaling molecule that can induce apoptosis or promote cellular survival depending on the context. Although not part of a specific protein complex 4-HNE can form adducts that alter the function of other proteins thereby affecting their biological roles.

Pathways

4-HNE is intertwined within oxidative stress and inflammation pathways affecting cellular redox balance and gene expression. It can upregulate antioxidants through the Nrf2 signaling pathway and inhibit pro-inflammatory pathways such as NF-kB. Related proteins such as glutathione S-transferases help detoxify 4-HNE by catalyzing the conjugation with glutathione decreasing its reactivity and toxicity.

Oxidative stress involving 4-Hydroxynonenal links to neurodegenerative diseases such as Alzheimer's disease and chronic inflammatory diseases like atherosclerosis. In Alzheimer's disease 4-HNE can modify amyloid-beta proteins increasing their aggregation and toxicity. In atherosclerosis 4-HNE modifies low-density lipoproteins (LDL) contributing to foam cell formation and plaque buildup. These modifications highlight 4-HNE's role in disease progression.
