Abcam's 8-iso-PGF2 alpha in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of 8-iso-PGF2 alpha in tissue culture media and urine.
A goat anti-rabbit IgG antibody has been precoated onto 96-well plates. Standards or test samples are added to the wells, along with an alkaline phosphatase (AP) conjugated-8-iso-PGF2 alpha antigen and a polyclonal rabbit antibody specific to 8-iso-PGF2 alpha. After incubation the excess reagents are washed away. pNpp substrate is added and after a short incubation the enzyme reaction is stopped and the yellow color generated is read at 405 nm. The intensity of the yellow coloration is inversely proportional to the amount of 8-iso-PGF2 alpha captured in the plate.
The measurement of F2-isoprostanes has emerged as one of the most reliable approaches to assess oxidative stress status in vivo, providing an important tool to explore the role of oxidative stress in the pathogenesis of human disease. This is a colorimetric competitive immunoassay kit for the quantitative determination of free 8-iso-Prostaglandin F2α in biological fluids such as urine and in culture supernatants. 8-iso-PGF2α is produced in vivo by both non-cyclooxygenase and cyclooxygenase dependant mechanisms from arachidonic acid. It has been shown to be a potent vasoconstrictor, a potential mediator of hepatorenal syndrome and a mutagen in 3T3 cells and vascular smooth muscle cells. 8-iso-PGF2α has been shown to circulate in plasma and is excreted in urine.

8-iso-PGF2α also known as 8-iso-PGF2a iso-8-PGF2 or PGF2 alpha is an isoprostane formed by the free-radical-catalyzed peroxidation of arachidonic acid. It has a mass of approximately 354 g/mol. This molecule is commonly found in cell membranes during oxidative stress. It exists in various tissues with higher expressions noted in the liver heart and brain. 8-iso-PGF2α can be detected using methods such as the Alpha ELISA or Iso Kit which are critical for measuring its levels in biological samples.
8-iso-PGF2α functions as a marker of oxidative stress within the organism. It participates in cell signaling affecting vascular tone and platelet aggregation. It does not form part of a larger protein complex by itself but acts through receptor-mediated mechanisms mainly interacting with thromboxane receptors to exert its effects.
