AKT 1/2/3 pS473 + AKT 1/2/3 Total (ab253299) is a single-wash 90 min sandwich ELISA designed for the semi-quantitative measurement of total AKT 1/2/3 pS473 + AKT 1/2/3 total protein in human and mouse cell lysates. It uses our proprietary SimpleStep ELISA® technology. Semi-quantitate AKT 1/2/3 pS473 + AKT 1/2/3 total with 1 μg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. AKT isoforms belong to a conserved family of serine/threonine protein kinases. AKT plays a central role in cell survival and inhibiting apoptosis, by phosphorylating and inactivating several targets. AKT is activated by insulin and various other agonists, which promote transport to the plasma membrane, where the activation loop of AKT is phosphorylated at Thr308 by PDK1. The carboxy terminus is phosphorylated at Ser473 by mammalian target of rapamycin complex 2 (TORC2).
AKT substrates include Bad, forkhead transcription factors, and caspase-9. Another vital AKT function is the regulation of glycogen synthesis through phosphorylation of GSK-3. AKT phosphorylation also prevents GSK-3β mediated phosphorylation and degradation of cyclin D1, and negatively regulates the cyclin dependent kinase inhibitors p27 Kip and p21 Waf1/CIP1. AKT also plays a critical role in the regulation of TORC1 and tuberin (TSC2).

Biological function summary
AKT proteins perform critical roles in key signaling pathways such as the PI3K/AKT pathway which is important for cellular processes like growth proliferation and survival. AKT activation requires phosphorylation at two sites: Thr308 in the activation loop and Ser473 in the hydrophobic motif. These kinases attend a complex signaling network involving direct phosphorylation of multiple downstream targets influencing numerous cellular processes. AKT1 AKT2 and AKT3 might have overlapping as well as unique functions in vivo reflecting their different tissue distributions and substrate specificities.
Pathways
AKT proteins are central components of the insulin signaling pathway and the PI3K/AKT/mTOR pathway. These pathways play essential roles in regulating metabolism cell survival and growth. AKT interacts with various other proteins such as mTOR and FOXO transcription factors. In the PI3K/AKT/mTOR pathway AKT acts downstream of PI3K to activate or inhibit targets that regulate protein synthesis metabolism and apoptosis.
