Product details

Abcam's Androstenedione in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Androstenedione in serum and plasma.

A 96-well plate has been precoated with anti-Androstenedione IgG. Samples and the Androstenedione-HRP conjugate are added to the wells, where Androstenedione in the sample competes with the added Androstenedione-HRP for antibody binding. After incubation, the wells are washed to remove unbound material and TMB substrate is then added which is catalyzed by HRP to produce blue coloration. The reaction is terminated by addition of Stop Solution which stops the color development and produces a color change from blue to yellow. The intensity of signal is inversely proportional to the amount of Androstenedione in the sample and the intensity is measured at 450 nm.

Androstenedione also known as 4-androstenedione is a steroid hormone with a molecular weight of approximately 290.44 g/mol. It serves as an important intermediate in the biosynthesis of androgens and estrogens. This hormone predominantly expresses in the adrenal glands ovaries and testes where it plays a significant role in the production of sex steroids. Androstenedione circulates in the blood and can be converted into testosterone and estrone by enzymes present in specific tissues.
Biological function summary

Androstenedione acts as a precursor for the synthesis of potent androgens and estrogens contributing to the development and maintenance of secondary sexual characteristics. It is not part of a larger protein complex operating instead through enzyme-mediated conversions. These include the aromatization to form estrone and the reduction to produce testosterone. These biochemical transformations influence physiological processes such as reproductive function and secondary sexual expression.

Pathways

Androstenedione participates in the steroidogenesis pathway which is essential for the production of steroid hormones. Cholesterol provides the initial substrate for this pathway which then leads to the formation of androstenedione through a series of enzymatic reactions. The cytochrome P450 enzyme family particularly CYP17A1 plays an important role in generating androstenedione from its precursors. Additionally it links to the estrogen biosynthesis pathway interacting with enzymes such as aromatase that convert androstenedione to estrone.

Androstenedione associates with conditions like polycystic ovary syndrome (PCOS) and adrenal hyperplasia. Both disorders involve dysregulation of steroid hormone production and result in elevated levels of androgenic hormones. In PCOS increased androstenedione levels contribute to symptoms such as hirsutism and infertility. Related proteins such as 17-beta-hydroxysteroid dehydrogenase type 3 also interact with androstenedione during the manifestation of these diseases. Understanding the role of androstenedione in these contexts aids in developing therapeutic strategies to manage hormone-related disorders.
