BDNF ELISA Kit- Intracellular is a single-wash 90-min Simplestep used to quantify BDNF with a sensitivity of 3.6pg/ml. The assay uses a simple mix-wash-read protocol with just one incubation and one wash step.

-Colormetric sandwich ELISA - 450 nm readout- works on any standard plate reader.

-Different formats for different needs: 10 x 96 plates for bulk orders and 384-well for higher throughput.

Interpolated concentration of native BDNF was measured in duplicate at different sample concentrations. Undiluted samples are 500 µg/mL human hippocampus tissue extract, mouse brain tissue extract, and rat cortex tissue extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer SSW.

Recombinant prepro-BDNF, pro-BDNF, and mature BDNF proteins were tested for reactivity in 1X Cell Extraction Buffer SSW. Background-subtracted data values (mean +/- SD) are graphed. The kit reacts with both mature and pro-BDNF but weakly detects prepro-BDNF.

How the assay works

BDNF ELISA Kit- Intracellular (ab324920) SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

Important signaling molecule that activates signaling cascades downstream of NTRK2 (By similarity). During development, promotes the survival and differentiation of selected neuronal populations of the peripheral and central nervous systems. Participates in axonal growth, pathfinding and in the modulation of dendritic growth and morphology. Major regulator of synaptic transmission and plasticity at adult synapses in many regions of the CNS. The versatility of BDNF is emphasized by its contribution to a range of adaptive neuronal responses including long-term potentiation (LTP), long-term depression (LTD), certain forms of short-term synaptic plasticity, as well as homeostatic regulation of intrinsic neuronal excitability (By similarity).. Neurotrophic factor BDNF precursor form. Important signaling molecule that activates signaling cascades downstream of NTRK2. Activates signaling cascades via the heterodimeric receptor formed by NGFR and SORCS2. Signaling via NGFR and SORCS2 plays a role in synaptic plasticity and long-term depression (LTD). Binding to NGFR and SORCS2 promotes neuronal apoptosis. Promotes neuronal growth cone collapse.
